产品介绍This gene encodes a cell surface glycoprotein that regulates complement-mediated cell lysis, and it is involved in lymphocyte signal transduction. This protein is a potent inhibitor of the complement membrane attack complex, whereby it binds complement C8 and/or C9 during the assembly of this complex, thereby inhibiting the incorporation of multiple copies of C9 into the complex, which is necessary for osmolytic pore formation. This protein also plays a role in signal transduction pathways in the activation of T cells. Mutations in this gene cause CD59 deficiency, a disease resulting in hemolytic anemia and thrombosis, and which causes cerebral infarction. Multiple alternatively spliced transcript variants, which encode the same protein, have been identified for this gene. [provided by RefSeq, Jul 2008]

CD5是67kDa跨膜糖蛋白，属于清道夫受体富含半胱氨酸超家族，主要表达于胸腺细胞、成熟T细胞、B-1细胞（如小鼠B1a细胞）及部分T细胞淋巴瘤/白血病细胞。它通过与CD72、TCR/BCR等分子相互作用，负向调节T/B细胞活化信号，在免疫耐受和自身免疫病中起关键作用。

实验应用关键要点

流式细胞术（T/B细胞亚群分析）：

样本处理：外周血单个核细胞（PBMC）用PBS+2%FBS洗涤，调整浓度至1×10⁶/mL；

染色条件：荧光标记抗体（如AF488偶联CD5）4℃避光孵育30分钟，无需透化（CD5为膜蛋白）；

设门策略：CD5⁺CD3⁺为T细胞群，CD5⁺CD19⁺可识别B-1细胞亚群。

IHC-P（淋巴瘤诊断）：

抗原修复：柠檬酸钠缓冲液（pH6.0） 微波修复15分钟，冷却至室温；

阳性定位：T细胞淋巴瘤（如 mantle cell lymphoma）胞膜呈棕黄色阳性，B细胞淋巴瘤（如滤泡性淋巴瘤）多为阴性；

对照设置：正常淋巴结组织（T细胞区阳性）和胃癌组织（阴性对照）。

WB实验（细胞裂解液检测）：

样本制备：Jurkat T细胞（高表达CD5）用含1% SDS的RIPA裂解液冰上裂解，12000g离心取上清；

电泳条件：10% SDS-PAGE凝胶，恒压120V分离，转膜后5%脱脂奶粉封闭1小时；

抗体孵育：一抗1:1000稀释（4℃过夜），HRP二抗1:5000室温孵育1小时，曝光30秒可见67kDa条带。
