产品介绍The protein encoded by this gene is actually a preproprotein that is cleaved into four distinct mature peptides. One of these, glucagon, is a pancreatic hormone that counteracts the glucose-lowering action of insulin by stimulating glycogenolysis and gluconeogenesis. Glucagon is a ligand for a specific G-protein linked receptor whose signalling pathway controls cell proliferation. Two of the other peptides are secreted from gut endocrine cells and promote nutrient absorption through distinct mechanisms. Finally, the fourth peptide is similar to glicentin, an active enteroglucagon. [provided by RefSeq].

靶点与功能

胰高血糖素（Glucagon）是由胰岛α细胞分泌的多肽激素，含29个氨基酸，分子量约3.5kDa，通过与G蛋白偶联受体结合，促进肝糖原分解和糖异生，升高血糖。其核心功能与研究价值包括：

代谢调控：与胰岛素共同维持血糖平衡，是2型糖尿病高血糖的重要驱动因子；

实验操作关键要点

IHC-P优化（胰腺组织切片）：

抗原修复：采用柠檬酸钠缓冲液（pH6.0） 室温修复30分钟（避免高温破坏多肽结构）；

阳性定位：胰岛α细胞胞质呈棕黄色颗粒状阳性，与胰岛素阳性β细胞呈“镶嵌分布”；

封闭处理：5%正常山羊血清+0.1% BSA室温封闭1小时，减少非特异性结合。

ELISA定量检测（血清样本）：

包被条件：抗体用0.05M碳酸盐缓冲液（pH9.6）稀释至2μg/mL，4℃包被过夜；

样本处理：血清1:10稀释（避免基质干扰），37℃孵育2小时；

显色控制：HRP标记二抗（1:5000）室温孵育1小时，TMB显色15分钟，450nm读取OD值（标准曲线范围0.1-10ng/mL）。
