产品介绍Modulation of the chromatin structure plays an important role in the regulation of transcription in eukaryotes. The nucleosome, made up of four core histone proteins (H2A, H2B, H3 and H4), is the primary building block of chromatin. The N-terminal tail of core histones undergoes different posttranslational modifications including acetylation, phosphorylation and methylation. These modifications occur in response to cell signal stimuli and have a direct effect on gene expression. In most species, the histone H2B is primarily acetylated at lysines 5, 12, 15 and 20. Histone H3 is primarily acetylated at lysines 9, 14, 18 and 23. Acetylation at lysine 9 appears to have a dominant role in histone deposition and chromatin assembly in some organisms. Phosphorylation at Ser10 of histone H3 is tightly correlated with chromosome condensation during both mitosis and meiosis.

靶点与功能

组蛋白H3是核小体核心组蛋白（H2A、H2B、H3、H4）之一，分子量约15kDa，含135个氨基酸，通过N端赖氨酸残基的甲基化、乙酰化等修饰参与染色质重塑与基因表达调控。作为内参的核心优势：

表达稳定性：在真核细胞中组成型高表达，不受细胞周期、营养状态或药物处理影响（除组蛋白去乙酰化酶抑制剂等特殊情况）；

核定位特异性：仅存在于细胞核，可通过核蛋白提取验证分离效果；

实验兼容性：适用于WB、IHC、IF等多种实验，是核蛋白定量的金标准内参。

实验操作关键要点

WB内参优化（核蛋白样本）：

样本处理：使用核蛋白提取试剂盒分离细胞核组分，BCA定量确保上样量一致（推荐20-30μg总蛋白）；

电泳条件：15% SDS-PAGE凝胶（因分子量小），恒压80V浓缩胶、120V分离胶，目标条带约15kDa；

抗体孵育：单抗1:1000稀释（5% BSA+TBST），4℃孵育过夜，HRP二抗（1:5000）室温孵育1小时，曝光时间5-10秒（避免过曝）。

IHC-P核定位验证：

抗原修复：采用柠檬酸钠缓冲液（pH6.0） 微波修复15分钟（95℃），确保核信号释放；

阳性定位：所有有核细胞细胞核呈均一棕黄色阳性，胞质无染色（可验证核蛋白提取纯度）；

对照设置：无需阴性对照（正常细胞均表达），但需确保组织固定充分（4%多聚甲醛固定12小时）。
