产品介绍Histones are basic nuclear proteins that are responsible for the nucleosome structure of the chromosomal fiber in eukaryotes. Nucleosomes consist of approximately 146 bp of DNA wrapped around a histone octamer composed of pairs of each of the four core histones (H2A, H2B, H3, and H4). The chromatin fiber is further compacted through the interaction of a linker histone, H1, with the DNA between the nucleosomes to form higher order chromatin structures. This gene is intronless and encodes a member of the histone H4 family. Transcripts from this gene lack polyA tails; instead, they contain a palindromic termination element. 靶点与功能

组蛋白H4是核心组蛋白家族成员（分子量约11.3kDa），与H2A、H2B、H3共同构成核小体核心颗粒，参与染色质结构维持和基因表达调控。其核心研究价值：

表观遗传标记：H4的赖氨酸乙酰化（如H4K5ac、H4K8ac）、甲基化（H4K20me3）等修饰与转录激活/沉默密切相关；

细胞周期调控：H4的磷酸化修饰在有丝分裂期染色体浓缩中起关键作用；

疾病标志物：H4修饰异常与肿瘤（如肺癌、前列腺癌）、神经退行性疾病相关。

实验操作关键要点

ChIP实验（染色质免疫沉淀）：

染色质制备：使用甲醛交联（1%甲醛室温固定10分钟），超声破碎获得200-500bp片段；

抗体用量：每10⁶细胞对应2-5μg抗体，4℃旋转孵育过夜；

对照设置：Input对照（未免疫沉淀的染色质）、阴性对照（IgG）。

WB检测（核蛋白提取物）：

样本处理：使用核蛋白提取试剂盒分离细胞核组分，避免胞质蛋白干扰；

上样量：核蛋白上样量5-10μg，15% SDS-PAGE凝胶分离（因H4分子量小）；

抗体孵育：一抗1:2000稀释（5% BSA+TBST），4℃孵育过夜，二抗1:5000室温孵育1小时。
