产品介绍The glycoprotein encoded by this gene is a cell surface antigen that is expressed in greater than 95% of human colon cancers. The open reading frame encodes a 319-amino acid polypeptide having a putative secretory signal sequence and 3 potential glycosylation sites. The predicted mature protein has a 213-amino acid extracellular region, a single transmembrane domain, and a 62-amino acid intracellular tail. The sequence of the extracellular region contains 2 domains characteristic of the CD2 subgroup of the immunoglobulin (Ig) superfamily. [provided by RefSeq, Jul 2008]

靶点与功能

33糖蛋白是ATP结合盒（ABC）转运蛋白超家族成员，分子量约170kDa，通过ATP水解能量将多种药物泵出细胞，是肿瘤多药耐药（MDR） 的关键介质。其核心研究价值包括：

肿瘤耐药：在乳腺癌、肺癌、卵巢癌等肿瘤中过表达，导致紫杉醇、阿霉素等化疗药物外排，降低细胞内药物浓度；

生理功能：正常组织（肝脏、肾脏、肠道）中参与外源性物质清除，保护细胞免受毒素损伤；

药物研发：作为逆转肿瘤耐药的靶点，用于筛选P-gp抑制剂（如维拉帕米）。

实验操作关键要点

IHC-P优化（肿瘤组织切片）：

抗原修复：采用EDTA缓冲液（pH9.0） 高压修复3分钟，增强细胞膜信号；

阳性定位：肿瘤细胞胞膜呈棕黄色线性阳性，部分胞质内可见颗粒状表达（药物外排活跃时）；

对照设置：必须包含P-gp高表达细胞系（如KB-V1） 和阴性对照（如LoVo细胞）。

WB检测注意事项：

样本处理：使用含1mM钒酸钠（磷酸酶抑制剂） 的RIPA裂解液冰上裂解30分钟，12000g离心取上清；

电泳条件：7.5% SDS-PAGE凝胶（因分子量较大），恒压80V浓缩胶、120V分离胶，转膜采用0.45μm PVDF膜（300mA恒流2小时）；

抗体孵育：重组兔单抗1:800稀释（5%脱脂奶粉+TBST），4℃摇床孵育过夜，HRP二抗（山羊抗兔）1:5000室温孵育1小时。
