产品介绍This gene encodes a 140 kDa protein that is stored in the alpha-granules of platelets and Weibel-Palade bodies of endothelial cells. This protein redistributes to the plasma membrane during platelet activation and degranulation and mediates the interaction of activated endothelial cells or platelets with leukocytes. The membrane protein is a calcium-dependent receptor that binds to sialylated forms of Lewis blood group carbohydrate antigens on neutrophils and monocytes. 靶点与功能

P选择素（CD62P）是钙依赖性粘附分子，平时“藏”在血小板的α颗粒和血管内皮细胞的Weibel-Palade小体里，当细胞受到刺激（比如炎症、凝血酶作用）时，会迅速“搬家”到细胞表面。它的主要作用是像“粘合剂”一样，介导白细胞（尤其是中性粒细胞、单核细胞）在血管内皮上的初始滚动，为后续的炎症细胞浸润和血栓形成“铺路”。在血栓性疾病、动脉粥样硬化、肿瘤转移等研究中，它都是重要的标志物和潜在治疗靶点。

实验应用

样本处理：如果是检测活化后的血小板或内皮细胞表面的P选择素，要注意样本制备时的刺激条件（比如用凝血酶刺激血小板），同时加入蛋白酶抑制剂防止蛋白降解。

IHC实验：在组织切片中，阳性信号主要在活化的血小板膜或血管内皮细胞表面，比如炎症组织中的血管内皮会显色，正常静息状态下表达较低，可设置阳性对照（如炎症模型组织）和阴性对照（如正常组织）。

WB实验：由于P选择素是糖蛋白，分子量较大且可能有糖基化修饰，WB结果中140kDa左右出现清晰条带即为阳性，上样量建议50-100μg总蛋白。
