产品介绍This gene encodes a member of the carcinoembryonic antigen (CEA) gene family, which belongs to the immunoglobulin superfamily. Two subgroups of the CEA family, the CEA cell adhesion molecules and the pregnancy-specific glycoproteins, are located within a 1.2 Mb cluster on the long arm of chromosome 19. Eleven pseudogenes of the CEA cell adhesion molecule subgroup are also found in the cluster. The encoded protein was originally described in bile ducts of liver as biliary glycoprotein. Subsequently, it was found to be a cell-cell adhesion molecule detected on leukocytes, epithelia, and endothelia. The encoded protein mediates cell adhesion via homophilic as well as heterophilic binding to other proteins of the subgroup. Multiple cellular activities have been attributed to the encoded protein, including roles in the differentiation and arrangement of tissue three-dimensional structure, angiogenesis, apoptosis, tumor suppression, metastasis, and the modulation of innate and adaptive immune responses. Multiple transcript variants encoding different isoforms have been reported, but the full-length nature of all variants has not been defined. [provided by RefSeq, May 2010].

靶点与功能

CEACAM1是免疫球蛋白超家族成员，分子量约140kDa（糖基化修饰后），通过同源/异源粘附参与细胞信号传导，其核心功能包括：

肿瘤调控：抑制肿瘤细胞增殖和转移（如结直肠癌、肺癌中高表达与预后正相关）；

免疫调节：作为免疫检查点分子，通过与T细胞表面受体结合抑制抗肿瘤免疫应答；

血管生成：调控内皮细胞粘附与血管形成，影响肿瘤微环境血供。

实验操作关键要点

IHC-P组织染色（肿瘤样本）：

抗原修复：采用柠檬酸钠缓冲液（pH6.0） 微波修复20分钟（95℃），增强细胞膜表面CEACAM1信号；

阳性定位：肿瘤细胞细胞膜/胞质呈棕黄色阳性，正常组织中多表达于上皮细胞；

对照设置：推荐使用CEACAM1高表达的结直肠癌细胞系（如HT-29）作为阳性对照。

WB验证（细胞裂解液）：

样本处理：使用含蛋白酶抑制剂（1mM PMSF） 的RIPA裂解液冰上裂解30分钟，12000g离心15分钟取上清；

电泳条件：8% SDS-PAGE凝胶（因分子量较大），恒压80V浓缩胶、120V分离胶，转膜选用0.45μm PVDF膜（300mA恒流2小时）。
