产品介绍This gene encodes a 110-kD transmembrane glycoprotein that is highly expressed by human monocytes and tissue macrophages. It is a member of the lysosomal/endosomal-associated membrane glycoprotein (LAMP) family. The protein primarily localizes to lysosomes and endosomes with a smaller fraction circulating to the cell surface. It is a type I integral membrane protein with a heavily glycosylated extracellular domain and binds to tissue- and organ-specific lectins or selectins. The protein is also a member of the scavenger receptor family. Scavenger receptors typically function to clear cellular debris, promote phagocytosis, and mediate the recruitment and activation of macrophages. Alternative splicing results in multiple transcripts encoding different isoforms. [provided by RefSeq, Jul 2008].

靶点与功能

CD68是溶酶体膜糖蛋白（分子量110kDa），主要表达于巨噬细胞、单核细胞、破骨细胞及部分树突状细胞，像“溶酶体的守门人”，参与细胞内吞、抗原呈递和炎症因子释放。在病理状态下，CD68⁺巨噬细胞浸润程度与动脉粥样硬化斑块稳定性、肿瘤免疫微环境（如TAM）、慢性炎症疾病密切相关，是评估免疫浸润的重要指标。

实验操作关键要点

IHC-P（肿瘤组织巨噬细胞检测）：

抗原修复：柠檬酸钠缓冲液（pH6.0）微波修复15分钟（95℃），冷却后用0.3% Triton X-100增强膜通透性；

阳性定位：巨噬细胞胞质呈棕黄色颗粒状（如肿瘤间质区的TAM），可与CD163（M2型巨噬细胞）共染区分亚型；

对照设置：淋巴结组织作阳性对照（窦组织细胞显色），肝组织作阴性对照（肝细胞不表达）。

WB验证（RAW264.7巨噬细胞）：

样本制备：RIPA裂解液（含蛋白酶抑制剂）冰上裂解30分钟，12000g离心取上清，上样量40μg总蛋白；

电泳条件：10% SDS-PAGE凝胶，转膜用0.45μm PVDF膜，5% BSA封闭1小时；

抗体孵育：一抗1:500稀释（4℃过夜），HRP二抗（山羊抗小鼠/兔）1:5000室温孵育1小时，曝光可见110kDa条带。

流式细胞术（外周血单核细胞）：

染色方案：FITC-CD68抗体与PE-CD14抗体（1:100）避光孵育20分钟（4℃），红细胞裂解后上机；

设门策略：CD14⁺CD68⁺为单核细胞群，正常占比约5%-10%。
