产品介绍This gene encodes a 110-kD transmembrane glycoprotein that is highly expressed by human monocytes and tissue macrophages. It is a member of the lysosomal/endosomal-associated membrane glycoprotein (LAMP) family. The protein primarily localizes to lysosomes and endosomes with a smaller fraction circulating to the cell surface. It is a type I integral membrane protein with a heavily glycosylated extracellular domain and binds to tissue- and organ-specific lectins or selectins. The protein is also a member of the scavenger receptor family. Scavenger receptors typically function to clear cellular debris, promote phagocytosis, and mediate the recruitment and activation of macrophages. Alternative splicing results in multiple transcripts encoding different isoforms. [provided by RefSeq, Jul 2008]

实验操作关键要点

IHC-P组织染色（石蜡切片）：

抗原修复：柠檬酸钠缓冲液（pH6.0）微波修复20分钟（95℃），冷却至室温后用3% H₂O₂阻断内源性过氧化物酶；

阳性定位：巨噬细胞胞质呈棕黄色颗粒状阳性（如肝脏库普弗细胞、肿瘤间质TAMs）；

对照设置：淋巴结组织（阳性对照，巨噬细胞丰富）和小肠上皮（阴性对照）。

WB验证（RAW264.7细胞裂解液）：

样本处理：含1% NP-40的RIPA裂解液冰上裂解30分钟，12000g离心15分钟取上清；

抗体稀释：重组兔单抗推荐1:5000稀释（5%脱脂奶粉+TBST），4℃孵育过夜，HRP二抗1:10000室温孵育1小时。
