产品介绍This gene encodes a 110-kD transmembrane glycoprotein that is highly expressed by human monocytes and tissue macrophages. It is a member of the lysosomal/endosomal-associated membrane glycoprotein (LAMP) family. The protein primarily localizes to lysosomes and endosomes with a smaller fraction circulating to the cell surface. It is a type I integral membrane protein with a heavily glycosylated extracellular domain and binds to tissue- and organ-specific lectins or selectins. The protein is also a member of the scavenger receptor family. Scavenger receptors typically function to clear cellular debris, promote phagocytosis, and mediate the recruitment and activation of macrophages. Alternative splicing results in multiple transcripts encoding different isoforms. [provided by RefSeq, Jul 2008]

实验操作关键要点

IHC实验（肿瘤组织切片）：

抗原修复：推荐EDTA缓冲液（pH8.0） 高温修复20分钟（95℃），因CD68糖基化表位对柠檬酸钠缓冲液敏感；

阳性定位：巨噬细胞胞质/溶酶体呈棕黄色颗粒状阳性，如肿瘤间质中的TAMs、炎症组织的巨噬细胞浸润区；

对照设置：用正常脾脏组织作阳性对照（红髓巨噬细胞强阳性），CD68敲除细胞系作阴性对照。

WB验证（RAW264.7巨噬细胞）：

样本处理：使用含1% NP-40的RIPA裂解液（避免破坏糖基化结构），冰上裂解30分钟，12000g离心取上清；

关键技巧：上样量需达80μg总蛋白，一抗1:1000稀释（5% BSA+TBST），4℃孵育过夜，曝光时间20-30秒（预期条带70-100kDa）。

流式细胞术（外周血单核细胞）：

抗体选择：优先用荧光标记抗体（如PE-CD68），避免二抗交叉反应；

染色步骤：细胞用含2% FBS的PBS重悬，抗体1:50稀释，4℃避光孵育30分钟，洗涤后上机（CD14⁺CD68⁺为单核巨噬细胞群）
