靶点与功能

CD7（也称GP40、LEU-9）是一种40kDa的I型跨膜糖蛋白，属于免疫球蛋白超家族，核心功能包括：

T细胞发育标记：在胸腺细胞和成熟T细胞表面高表达，是T细胞早期活化的关键标志物；

免疫细胞分型：表达于NK细胞和部分造血干细胞，可用于免疫细胞亚群鉴定；

临床关联：在T细胞白血病、淋巴瘤中异常高表达，是白血病免疫分型的重要指标。

实验操作关键要点

流式细胞术（FC）优化：

细胞制备：新鲜外周血或骨髓单细胞悬液经0.2% BSA-PBS洗涤2次，避免使用EDTA（影响受体构象）；

抗体孵育：1μg抗体/1×10⁶细胞，4℃避光孵育30分钟，流式缓冲液洗涤后上机，推荐与CD3/CD56抗体联合标记，区分T细胞（CD3⁺CD7⁺）和NK细胞（CD3⁻CD56⁺CD7⁺）。

WB检测注意事项：

样本处理：T细胞/白血病细胞裂解液需添加10mM DTT（破坏二硫键，避免多聚体干扰），冰上裂解30分钟，12000g离心15分钟取上清；

电泳条件：12% SDS-PAGE凝胶，恒压80V浓缩胶、120V分离胶，转膜选用0.45μm PVDF膜（250mA恒流1.5小时）。
