产品介绍This gene encodes a member of the heat shock protein 70 family, which contains both heat-inducible and constitutively expressed members. This protein belongs to the latter group, which are also referred to as heat-shock cognate proteins. It functions as a chaperone, and binds to nascent polypeptides to facilitate correct folding. It also functions as an ATPase in the disassembly of clathrin-coated vesicles during transport of membrane components through the cell. Alternatively spliced transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Aug 2011]

靶点与功能

HSP71（HSC70/HSPA8）是热休克蛋白70家族成员（分子量约71kDa），属于组成型表达的分子伴侣，核心功能包括：

蛋白稳态调控：通过ATP依赖的方式辅助新生肽链折叠、错误折叠蛋白降解，维持细胞内蛋白平衡；

应激保护：在热应激、氧化损伤等条件下表达上调，保护细胞免受凋亡；

疾病关联：在肿瘤（如肺癌、乳腺癌）中高表达，与细胞增殖、耐药相关；在神经退行性疾病（如阿尔茨海默病）中参与淀粉样蛋白清除。

实验操作关键要点

WB检测（细胞裂解液）：

样本处理：使用含1mM ATP的RIPA裂解液（维持HSP71活性）冰上裂解30分钟，12000g离心15分钟取上清；

电泳条件：10% SDS-PAGE凝胶，恒压80V浓缩胶、120V分离胶，转膜用0.45μm PVDF膜（250mA恒流1.5小时）；

抗体孵育：小鼠单抗1:8000稀释（5%脱脂奶粉+TBST），4℃孵育过夜，HRP二抗（山羊抗小鼠）1:5000室温孵育1小时。

IHC-P组织染色（肿瘤样本）：

抗原修复：采用柠檬酸钠缓冲液（pH6.0） 微波修复20分钟（95℃），冷却后用PBS洗3次；

阳性对照：推荐使用热应激处理的HepG2细胞爬片（胞质强阳性）；

显色控制：DAB显色5-8分钟，阳性信号为棕黄色颗粒（肿瘤细胞胞质弥漫性表达）。
