产品介绍The protein encoded by this gene associates with class II major histocompatibility complex (MHC) and is an important chaperone that regulates antigen presentation for immune response. It also serves as cell surface receptor for the cytokine macrophage migration inhibitory factor (MIF) which, when bound to the encoded protein, initiates survival pathways and cell proliferation. This protein also interacts with amyloid precursor protein (APP) and suppresses the production of amyloid beta (Abeta). Multiple alternatively spliced transcript variants encoding different isoforms have been identified. [provided by RefSeq, Aug 2011]

靶点与功能

CD74（ invariant chain，Ii链）是一种II型跨膜蛋白，像“分子伴侣”一样辅助MHC II类分子的折叠与转运，同时也是巨噬细胞迁移抑制因子（MIF）的受体。它的核心功能包括：

免疫调节：在B细胞、树突状细胞（DC）表面高表达，参与抗原提呈和T细胞活化；

肿瘤关联：在B细胞淋巴瘤（如弥漫大B细胞淋巴瘤）、多发性骨髓瘤中异常过表达，成为免疫治疗靶点；

信号通路：通过MIF/CD74轴激活AMPK-FOXO3a等通路，调控细胞增殖与抗凋亡（文献支持：Springer, 2025）。

实验应用关键操作

WB检测（Raji B淋巴瘤细胞）：

样本处理：RIPA裂解液（含1% NP-40）冰上裂解30分钟，12000g离心取上清，上样量50μg总蛋白；

关键技巧：CD74分子量约41kDa，使用12% SDS-PAGE凝胶，一抗1:800稀释（5% BSA+TBST），4℃孵育过夜，曝光可见单一清晰条带。

IHC诊断（淋巴瘤组织切片）：

抗原修复：柠檬酸钠缓冲液（pH6.0）微波修复15分钟，3% H₂O₂阻断内源性过氧化物酶；

阳性定位：B细胞淋巴瘤胞膜/胞质呈棕黄色阳性，正常淋巴组织中B细胞区（如生发中心）弱阳性；

对照设置：用扁桃体组织作阳性对照，CD74敲除细胞系作阴性对照。

功能实验（抗体依赖性细胞毒性，ADCC）：

效应细胞：人外周血单个核细胞（PBMC）；

靶细胞：CD74⁺ Raji细胞（CFSE标记）；

实验分组：（10μg/mL）vs IgG对照组，4小时后流式检测靶细胞死亡率（Annexin V⁺PI⁺比例）。
