产品介绍CD74, also known as the MHC class II associated invariant chain (Ii), is a type II transmembrane protein which binds to the peptide binding groove of newly synthesized MHC class II alpha/beta heterodimers and prevents their premature association with endogenous polypeptides. CD74 is produced in molar excess over MHC class II and some of this is expressed by an unknown pathway on the cell surface independent of, or in association with, MHC class II molecules. The half life of CD74 on the cell surface is only 3 to 4 min after which it is internalized. It has been proposed that cell surface CD74 binds The protein encoded by this gene associates with class II major histocompatibility complex (MHC) and is an important chaperone that regulates antigen presentation for immune response. It also serves as cell surface receptor for the cytokine macrophage migration inhibitory factor (MIF) which, when bound to the encoded protein, initiates survival pathways and cell proliferation. This protein also interacts with amyloid precursor protein (APP) and suppresses the production of amyloid beta (Abeta). 靶点与功能

CD74（又称MHCⅡ类分子不变链Ii）是一种25kDa的Ⅱ型跨膜糖蛋白，核心功能包括：

MHCⅡ类分子伴侣：辅助MHCⅡ类分子在内质网中折叠组装，阻止其提前结合自身抗原肽；

抗原肽加载调控：在内体中被蛋白酶水解生成CLIP片段（Ⅱ类分子相关 invariant链肽），为外源性抗原肽结合提供“占位保护”；

肿瘤免疫调节：在B细胞淋巴瘤、胰腺癌等肿瘤中高表达，通过与巨噬细胞表面受体结合促进免疫抑制性微环境形成。

实验操作关键要点

WB检测（抗原呈递细胞样本）：

样本处理：树突状细胞/巨噬细胞裂解液需添加1mM PMSF+10mM EDTA（抑制组织蛋白酶对CD74的降解），冰上裂解30分钟；

电泳条件：12% SDS-PAGE凝胶，恒压80V浓缩胶、120V分离胶，转膜选用0.22μm PVDF膜（增强小分子蛋白吸附）；

抗体孵育：一抗推荐1:1000稀释（5%脱脂奶粉+TBST），4℃孵育过夜，HRP二抗（山羊抗兔）1:5000室温孵育1小时。

IF共定位实验：

推荐组合：与MHCⅡ类分子抗体（如HLA-DR） 双标，Alexa Fluor 488标记CD74（绿色，内体定位），Alexa Fluor 594标记MHCⅡ（红色，细胞膜/内体），激光共聚焦观察二者在抗原提呈细胞内体中的共定位。
