Surface Ig non-covalently associates with two other proteins, Ig-alpha and Ig-beta, which are necessary for expression and function of the B-cell antigen receptor. 靶点与功能

CD79A（Igα）是B细胞抗原受体（BCR）复合物的关键亚基，与CD79B（Igβ）以异二聚体形式存在，像“信号转换器”一样，将B细胞表面抗体识别的抗原信号传递到细胞内，启动B细胞活化、增殖和抗体分泌。它的表达贯穿B细胞发育全过程（从祖B细胞到成熟B细胞），但在浆细胞中下调，因此是：

B细胞淋巴瘤病理诊断的金标准标志物（如弥漫大B细胞淋巴瘤、滤泡性淋巴瘤）；

B细胞发育分化研究的关键工具（区分B细胞与T细胞、髓系细胞）。

实验应用关键操作

IHC病理诊断（淋巴瘤组织切片）：

抗原修复：柠檬酸钠缓冲液（pH6.0）高温高压修复2分钟，自然冷却后3% H₂O₂阻断内源性过氧化物酶；

阳性定位：B细胞胞膜/胞质呈棕黄色阳性，淋巴瘤组织中阳性细胞比例＞90%，可与CD20联合染色（CD20⁺CD79A⁺为成熟B细胞淋巴瘤）；

对照设置：用扁桃体组织作阳性对照（生发中心B细胞强阳性），T细胞淋巴瘤组织作阴性对照（无信号）。

WB验证（Raji B淋巴瘤细胞系）：

样本处理：RIPA裂解液（含1mM PMSF）冰上裂解30分钟，12000g离心取上清，上样量30μg总蛋白；

关键技巧：使用12% SDS-PAGE凝胶（分离小分子量蛋白），一抗1:1000稀释（5% BSA+TBST），4℃孵育过夜，曝光10-15秒可见25-30kDa条带。

流式细胞术（外周血B细胞检测）：

染色方案：FITC标记CD79A抗体与PE-CD19抗体（1:100）避光孵育20分钟（4℃），红细胞裂解后上机；

设门策略：CD19⁺CD79A⁺为B细胞群，正常外周血中占比约5%-15%。
