产品介绍The protein encoded by this gene is a cell surface glycoprotein involved in leukocyte migration, T-cell adhesion, ganglioside GM1 and transmembrane protein transport, and T-cell death by a caspase-independent pathway. In addition, the encoded protein may have the ability to rearrange the actin cytoskeleton and may also act as an oncosuppressor in osteosarcoma. This gene is found in the pseudoautosomal region of chromosomes X and Y and escapes X-chromosome inactivation. There is a related pseudogene located immediately adjacent to this locus. 靶点与功能

CD99（MIC2）是跨膜糖蛋白，像“细胞黏附信号开关”一样，参与T细胞黏附、白细胞迁移和细胞凋亡调控。它的核心研究价值在于：

肿瘤诊断：尤因肉瘤/原始神经外胚层肿瘤（ES/PNET）的特异性标志物（阳性率＞95%），免疫组化染色是病理鉴别诊断的“金标准”；

淋巴瘤分型：在T淋巴母细胞淋巴瘤中高表达，可与CD3联合染色区分T/B细胞来源；

实体瘤研究：肝细胞癌、胃癌中CD99高表达提示转移风险增加，是潜在预后评估指标。

实验应用关键操作

IHC病理诊断（尤因肉瘤组织切片）：

抗原修复：柠檬酸钠缓冲液（pH6.0）高温高压修复2分钟，自然冷却后3% H₂O₂阻断内源性过氧化物酶；

阳性定位：肿瘤细胞胞膜呈棕黄色线性阳性，与CD99阴性的神经母细胞瘤、横纹肌肉瘤鉴别（阳性率差异＞90%）；

对照设置：用正常胸腺组织作阳性对照（皮质T细胞弱阳性），CD99敲除细胞系作阴性对照。

WB验证（SK-N-MC尤因肉瘤细胞系）：

样本处理：RIPA裂解液（含1% NP-40）冰上裂解30分钟，12000g离心取上清，上样量50μg总蛋白；

关键技巧：使用15% SDS-PAGE凝胶（分离小分子量蛋白），一抗1:1000稀释（5% BSA+TBST），4℃孵育过夜，曝光15-20秒可见16-20kDa条带。

流式细胞术（淋巴瘤细胞检测）：

染色方案：PE标记CD99抗体与FITC-CD3抗体（1:100）避光孵育20分钟（4℃），红细胞裂解后上机；

设门策略：CD3⁺CD99⁺为T淋巴母细胞淋巴瘤细胞群，正常外周血T细胞CD99弱表达。
