产品介绍Monomeric IgA constitutes 5-15 % of the serum immunoglobulins whereas dimeric IgA is localized to mucosa surfaces such as saliva, gastrointestinal secretion, bronchial fluids and milk. Mucosal IgA plays a major role in host defence by neutralising infectious agents at mucosal surfaces. The production is usually local and antigen specific IgA producing B-cells can be found in regions under the lamina propria where they mature into dimeric IgA producing plasma cells. IgA deficiency is the most common immunodeficiency that may affect both serum and mucosal produced IgA.

靶点与功能

血清型免疫球蛋白A（Serum IgA）是由浆细胞合成的糖蛋白，分子量约160kDa，以单体形式存在（占血清总Ig的10%-15%）。其核心功能包括：

免疫防御：通过中和病原体（如病毒、细菌）、激活补体替代途径参与体液免疫；

临床意义：血清IgA水平异常与IgA肾病（系膜区沉积）、自身免疫病、慢性感染（结核病）及免疫缺陷病（选择性IgA缺乏症）密切相关；

研究价值：作为黏膜免疫的“血清标志物”，可辅助分析全身免疫状态与局部黏膜免疫的关联。

实验操作关键要点

ELISA定量检测（血清样本）：

包被条件：兔多抗用0.05M碳酸盐缓冲液（pH9.6）稀释至2μg/mL，4℃包被过夜；

样本处理：血清需1:100稀释（含1% BSA的PBS），37℃孵育1小时（避免溶血样本干扰）；

显色优化：HRP标记二抗（山羊抗兔）1:5000稀释，TMB显色10分钟，450nm读取OD值（标准曲线范围0.1-100ng/mL）。

WB验证（血清或细胞裂解液）：

样本制备：血清直接上样（1-5μL/孔），或浆细胞裂解液（含蛋白酶抑制剂）12000g离心取上清；

电泳条件：10% SDS-PAGE凝胶，恒压80V浓缩胶、120V分离胶，转膜使用0.45μm硝酸纤维素膜；

抗体孵育：一抗1:500稀释（5%脱脂奶粉+TBST），4℃孵育过夜，二抗1:5000室温孵育1小时，曝光30-60秒（目标条带约160kDa）。
