产品介绍IgM normally constitutes about 10% of serum immunoglobulins. IgM antibody is prominent in early immune responses to most antigens and is largely confined to plasma due to it's large size. Monomeric IgM is expressed as a membrane bound antibody on the surface of B cells and as a pentamer when secreted by plasma cells. Due to it's high valency IgM is more efficient than other isotypes is binding antigens with repeating epitopes (virus particles and red blood cells) and is more efficient than IgG in activiating the complement pathway. The gene for the mu constant region contains four domains separated by short intervening sequences.

靶点与功能

人IgM是五聚体结构的免疫球蛋白（分子量约970kDa，含J链），主要由未成熟B细胞和活化B细胞分泌，核心研究价值：

早期免疫应答：感染早期（如病毒、细菌）最先产生，是急性感染的重要标志物；

B细胞发育：未成熟B细胞表面表达膜结合型IgM（mIgM），参与BCR信号通路；

疾病关联：在原发性胆汁性胆管炎（PBC）中抗线粒体IgM抗体阳性，在冷球蛋白血症中形成IgM免疫复合物。

实验操作关键要点

ELISA检测（血清IgM定量）：

包被条件：抗体用碳酸盐缓冲液（pH9.6）稀释至2μg/mL，4℃包被过夜；

样本处理：血清1:1000稀释（避免Hook效应），37℃孵育1小时；

显色控制：HRP标记二抗（山羊抗小鼠/兔）1:5000稀释，TMB显色15分钟，450nm读取OD值（标准曲线范围0.1-10μg/mL）。

流式细胞术（B细胞表面mIgM检测）：

样本处理：外周血单个核细胞（PBMC）用含2% FBS的PBS重悬（1×10⁶ cells/mL）；

抗体孵育：FITC标记IgM抗体1:100稀释，4℃避光孵育30分钟，洗涤后上流式仪；

对照设置：同型对照（小鼠/兔IgG-FITC）、阳性对照（CD19⁺ B细胞群体）。
