产品介绍All five immunoglobulin classes share the same basic four polypeptide chain structure of two heavy-chains and two light chains. There are five heavy chain types, and two light-chain types (Kappa and Lambda) both having a molecular weight of 22.5kDa. Any heavy-chain type can associate with either light-chain type, but on any immunoglobulin molecule both light-chains are of the same type. Kappa and Lambda consist of a variable region and a constant region and can easily be differentiated by the antigenic properties of the constant region.

λ轻链（Lambda Light Chain）是免疫球蛋白（Ig）的组成部分，与重链通过二硫键连接，分子量约25kDa，含恒定区（Cλ） 和可变区（Vλ）。其核心功能与研究价值包括：

B细胞克隆性鉴定：在淋巴瘤诊断中，λ轻链限制性表达（λ⁺细胞比例>90%）提示单克隆增生，是区分良恶性B细胞增殖的金标准；

免疫球蛋白分型：人类血清Ig中λ轻链占比约40%，与κ轻链（60%）的比值异常（如λ/κ>2或<0.5）提示浆细胞病（如多发性骨髓瘤）；

疾病标志物：游离λ轻链（FLC）升高是轻链型淀粉样变性的特异性指标，抗体可用于血清FLC定量检测。

实验操作关键要点

IHC-P淋巴瘤诊断（石蜡切片）：

抗原修复：采用柠檬酸钠缓冲液（pH6.0） 微波修复20分钟（95℃），增强胞质信号；

阳性定位：B细胞/浆细胞胞质呈棕黄色颗粒状阳性，λ轻链限制性表达时肿瘤细胞弥漫强阳性；

对照设置：必须包含反应性淋巴结炎组织（κ/λ混合阳性）和λ型多发性骨髓瘤组织（单一λ阳性）。

流式细胞术检测（外周血B细胞）：

样本处理：EDTA抗凝全血经红细胞裂解液处理后，用含2% FBS的PBS重悬至1×10⁶细胞/mL；

抗体组合：λ轻链抗体（FITC标记）+κ轻链抗体（PE标记）+CD19抗体（APC标记），4℃避光孵育30分钟；

阳性判定：正常B细胞κ⁺/λ⁺比例约6:4，单克隆B细胞群仅表达κ或λ（如λ⁺细胞>95%）。
