产品介绍Interleukin 2 (IL2) is a secreted cytokine that is important for the proliferation of T and B lymphocytes. The receptor of this cytokine is a heterotrimeric protein complex whose gamma chain is also shared by interleukin 4 (IL4) and interleukin 7 (IL7). The expression of this gene in mature thymocytes is monoallelic, which represents an unusual regulatory mode for controlling the precise expression of a single gene. The targeted disruption of a similar gene in mice leads to ulcerative colitis like disease, which suggests an essential role of this gene in the immune response to antigenic stimuli. IL2 has been shown to have antitumor effects in some studies. This is probably mediated by cytotoxic effector cells.

靶点与功能

IL-2是T细胞来源的多效性细胞因子（分子量约15kDa），通过与IL-2受体（IL-2Rα/β/γ）结合，调控免疫细胞命运：

免疫激活：促进CD4⁺ T细胞分化为效应T细胞（Th1/Th2）和调节性T细胞（Treg）；

抗肿瘤作用：IL-2是**获批的免疫治疗药物（Aldesleukin），用于转移性肾癌和黑色素瘤；

疾病关联：IL-2表达异常与自身免疫病（如多发性硬化）、免疫缺陷病相关。

实验操作关键要点

ELISA检测（血清IL-2定量）：

样本处理：血清需4℃离心（3000g×10分钟） 去除沉淀，避免溶血样本干扰；

包被条件：抗体用0.05M碳酸盐缓冲液（pH9.6）稀释至2μg/mL，4℃包被16小时；

显色控制：HRP标记二抗（1:5000）37℃孵育1小时，TMB显色10分钟，450nm读取OD值（标准曲线范围50-1000pg/mL）。

WB验证（活化T细胞裂解液）：

样本制备：PHA刺激的PBMC用含1mM PMSF的RIPA裂解液冰上裂解30分钟，12000g离心取上清；

电泳条件：15% SDS-PAGE凝胶（因IL-2分子量小），恒压80V浓缩胶、120V分离胶，转膜采用0.22μm PVDF膜；

抗体孵育：一抗1:1000稀释（5% BSA+TBST），4℃孵育过夜，曝光时间30-60秒（目标条带15kDa）。
