产品介绍This gene encodes a member of the TGF-beta (transforming growth factor-beta) superfamily of proteins. The encoded preproprotein is proteolytically processed to generate multiple peptide products, including the alpha subunit of the inhibin A and B protein complexes. These complexes negatively regulate follicle stimulating hormone secretion from the pituitary gland. Inhibins have also been implicated in regulating numerous cellular processes including cell proliferation, apoptosis, immune response and hormone secretion. Mutations in this gene may be associated with male infertility and premature ovarian failure in female human patients. [provided by RefSeq, Aug 2016]

抑制素α（Inhibin α）是二聚体糖蛋白激素的共有亚单位，分子量约51-62kDa（含糖基化修饰），需与βA亚单位结合形成抑制素A（Inhibin A），或与βB亚单位形成抑制素B。其核心功能包括：

内分泌调控：通过负反馈抑制垂体促卵泡生成素（FSH） 分泌，参与卵泡发育、卵巢周期及妊娠维持；

肿瘤关联：在卵巢颗粒细胞瘤、睾丸支持细胞瘤中高表达，是性索间质肿瘤诊断的特异性标志物；

妊娠监测：血清抑制素A水平与先兆子痫、胎儿染色体异常（如唐氏综合征）相关，可作为妊娠相关疾病的辅助诊断指标。

实验操作关键要点

IHC-P组织染色（卵巢/睾丸肿瘤样本）：

抗原修复：采用EDTA缓冲液（pH9.0） 高压修复3分钟（针对福尔马林固定石蜡切片），增强细胞膜及胞质信号；

阳性定位：卵巢颗粒细胞瘤中胞质呈棕黄色颗粒状阳性，正常卵巢组织中弱阳性或阴性；

对照设置：需包含卵巢颗粒细胞瘤组织（阳性对照）和子宫平滑肌组织（阴性对照）。

ELISA定量检测（血清/培养液样本）：

包被优化：抗体用0.05M碳酸盐缓冲液（pH9.6）稀释至2μg/mL，4℃包被过夜；

样本处理：血清1:10稀释（含1% BSA的PBS），37℃孵育1小时，避免高脂样本干扰；

显色控制：HRP标记二抗（山羊抗兔IgG）1:5000稀释，TMB显色15分钟，450nm读取OD值（标准曲线范围0.1-200pg/mL）。
