产品介绍This gene encodes a ubiquitous nuclear protein that stimulates both cell proliferation and differentiation. It is a member of the MADS (MCM1, Agamous, Deficiens, and SRF) box superfamily of transcription factors. This protein binds to the serum response element (SRE) in the promoter region of target genes. This protein regulates the activity of many immediate-early genes, for example c-fos, and thereby participates in cell cycle regulation, apoptosis, cell growth, and cell differentiation. This gene is the downstream target of many pathways; for example, the mitogen-activated protein kinase pathway (MAPK) that acts through the ternary complex factors (TCFs). 靶点与功能

STAT3（Signal Transducer and Activator of Transcription 3）是潜伏型细胞质转录因子，在细胞因子（如IL-6、IL-10）、生长因子（EGF、HGF）刺激下，通过Tyr705位点磷酸化形成同源二聚体，或与Ser727位点磷酸化协同调控下游靶基因表达。其核心功能与疾病关联包括：

肿瘤进展：激活后上调Cyclin D1、Bcl-2等基因，促进细胞增殖并抑制凋亡，在肝癌、乳腺癌中p-STAT3（Tyr705）阳性率＞60%（临床样本数据）；

炎症调控：通过IL-6/STAT3轴参与慢性炎症向癌症转化，抑制M1型巨噬细胞极化；

治疗靶点：p-STAT3抑制剂（如Stattic）已进入临床Ⅱ期试验，抗体检测可用于药效评估。

实验操作关键要点

WB检测（细胞信号通路激活验证）：

样本处理：HeLa细胞用IL-6（10ng/mL）刺激30分钟，RIPA裂解液（含磷酸酶抑制剂PhosSTOP）冰上裂解20分钟，12000g离心取上清；

电泳条件：8% SDS-PAGE凝胶，恒压80V浓缩胶、120V分离胶，转膜采用0.45μm PVDF膜（300mA恒流2小时）；

抗体孵育：一抗1:1000稀释（5% BSA+TBST），4℃孵育过夜，HRP二抗（山羊抗兔）1:5000室温孵育1小时，曝光时间30-60秒（需同时检测总STAT3作为内参）。

IHC-P组织染色（肿瘤样本）：

抗原修复：柠檬酸盐缓冲液（pH6.0）微波修复（高火5min→停火3min→中火5min），自然冷却至室温；

阳性定位：肿瘤细胞核呈棕黄色阳性（Tyr705磷酸化入核），细胞质弱阳性（Ser727磷酸化）；

封闭处理：5%山羊血清+0.3% Triton X-100室温封闭1小时，减少非特异性结合。

IF双标实验（信号共定位）：

推荐与p-ERK1/2抗体共染，使用Alexa Fluor 488（绿色，p-STAT3 Tyr705）和594（红色，p-ERK1/2），激光共聚焦观察细胞因子刺激后二者在核内共定位。
