产品介绍There are at least four distinct but related alkaline phosphatases: intestinal, placental, placental-like, and liver/bone/kidney (tissue non-specific). The first three are located together on chromosome 2 while the tissue non-specific form is located on chromosome 1. The product of this gene is a membrane bound glycosylated enzyme, also referred to as the heat stable form, that is expressed primarily in the placenta although it is closely related to the intestinal form of the enzyme as well as to the placental-like form. The coding sequence for this form of alkaline phosphatase is unique in that the 3' untranslated region contains multiple copies of an Alu family repeat. In addition, this gene is polymorphic and three common alleles (type 1, type 2 and type 3) for this form of alkaline phosphatase have been well characterized. [provided by RefSeq, Jul 2008]

靶点与功能

人胎盘泌乳素（Human Placental Lactogen, HPL）又称绒毛膜生长催乳素（hCS），是由胎盘合体滋养层细胞合成的单链多肽激素（191个氨基酸，分子量约22kDa）。其核心功能与临床意义包括：

妊娠维持：促进乳腺发育为泌乳做准备，增强母体对胎儿的营养供应（通过增加游离脂肪酸释放、减少母体葡萄糖利用）；

胎盘功能评估：血清HPL水平随妊娠进展升高（妊娠34-36周达高峰，正常范围4-11mg/L），分娩后24小时内迅速下降，可作为胎盘功能不全的早期预警指标（如胎儿生长受限、子痫前期）；

疾病关联：在葡萄胎、绒毛膜癌等滋养细胞疾病中异常高表达，是鉴别诊断和疗效监测的生物标志物。

实验操作关键要点

ELISA定量检测（血清样本）：

包被优化：多克隆抗体用碳酸盐缓冲液（pH9.6）稀释至2μg/mL，4℃包被过夜；单克隆抗体推荐配对使用（捕获抗体识别N端，检测抗体识别C端）以提高特异性；

样本处理：血清需1:100稀释（含1% BSA的PBS），37℃孵育1小时，避免溶血样本干扰（血红蛋白会降低HPL稳定性）；

信号检测：HRP标记二抗1:8000稀释，TMB显色15分钟，450nm读取OD值（标准曲线范围0.1-50ng/mL，R²需≥0.99）。

IHC-P组织定位（胎盘样本）：

抗原修复：柠檬酸盐缓冲液（pH6.0）微波修复15分钟（石蜡切片），增强滋养细胞胞质信号；

阳性定位：正常胎盘组织合体滋养细胞胞质呈棕黄色阳性，细胞滋养细胞弱阳性；

对照设置：推荐使用妊娠中期胎盘组织（强阳性对照）和非妊娠子宫组织（阴性对照）。

WB验证（胎盘组织裂解液）：

样本制备：胎盘绒毛组织用含蛋白酶抑制剂（1mM PMSF） 的RIPA裂解液冰上裂解30分钟，12000g离心取上清；

电泳条件：12% SDS-PAGE凝胶，恒压100V电泳90分钟，转PVDF膜（0.22μm）；

抗体孵育：一抗1:1000稀释（5%脱脂奶粉+TBST），4℃孵育过夜，曝光时间30秒（条带位于22kDa处，无明显杂带）。
