产品介绍Placental lactogen is a member of the somatotropin/prolactin family of hormones and plays an important role in growth control. The gene is located at the growth hormone locus on chromosome 17 along with four other related genes in the same transcriptional orientation; an arrangement which is thought to have evolved by a series of gene duplications. Although the five genes share a remarkably high degree of sequence identity, they are expressed selectively in different tissues. Alternative splicing generates additional isoforms of each of the five growth hormones, leading to further diversity and potential for specialization. This particular family member is expressed mainly in the placenta and utilizes multiple transcription initiation sites.

人胎盘泌乳素（Human Placental Lactogen, HPL，又称绒毛膜生长催乳素）是由胎盘滋养层合体细胞分泌的多肽激素（217个氨基酸，分子量约22kDa），与生长激素（GH）和泌乳素（PRL）同属GH/PRL家族。其核心功能与临床意义包括：

妊娠维持：促进母体乳腺发育及脂解代谢，为胎儿提供游离脂肪酸；妊娠第6周开始分泌，34-36周达峰值（血清浓度5-15μg/mL），是胎盘功能不全的敏感指标（如胎儿生长受限FGR时HPL水平降低30%以上）；

肿瘤关联：在滋养细胞肿瘤（如葡萄胎、绒毛膜癌）中异常高表达，可作为疗效监测标志物；近年发现其通过激活PRLR-STAT5通路促进乳腺癌细胞增殖（Cancer Res, 2025）；

独特信号机制：不结合生长激素受体（GHR），而是通过锌离子诱导的PRLR二聚化激活下游信号（与PRL共享受体但亲和力低10倍）。

实验操作关键要点

ELISA定量检测（血清/胎盘组织匀浆）：

包被优化：抗体用碳酸盐缓冲液（pH9.6）稀释至1μg/mL，4℃包被过夜，PBST洗涤3次（每次5分钟）；

样本处理：血清需1:100稀释（含1% BSA的PBS），胎盘组织匀浆需用含1mM PMSF的RIPA裂解液制备，37℃孵育2小时；

信号放大：HRP标记二抗（山羊抗兔）1:8000稀释，TMB显色15分钟，450nm读取OD值（标准曲线范围0.1-50ng/mL，R²需≥0.99）。

IHC-P胎盘组织染色：

抗原修复：采用柠檬酸盐缓冲液（pH6.0） 微波修复20分钟（95℃），滋养层细胞胞质信号显著增强；

阳性定位：正常胎盘组织合体滋养细胞层呈棕黄色颗粒状阳性，胎儿生长受限（FGR）样本中阳性细胞比例降低（<30%）；

对照设置：必须包含妊娠中期胎盘组织（强阳性）和非妊娠子宫肌层（阴性对照）。

WB验证（JEG-3细胞裂解液）：

样本制备：人绒毛膜癌细胞JEG-3用含磷酸酶抑制剂的RIPA裂解液冰上裂解30分钟，12000g离心取上清；

电泳条件：12% SDS-PAGE凝胶，恒压80V浓缩胶、120V分离胶，转膜使用0.22μm PVDF膜（250mA恒流1小时）；

抗体孵育：一抗1:1500稀释（5% BSA+TBST），4℃孵育过夜，曝光时间45秒（避免与25kDa附近杂蛋白混淆）。
