产品介绍Insulin is one of the major regulatory hormones of intermediate metabolism throughout the body. The biological actions of this hormone involve integration of carbohydrate, protein, and lipid metabolism. Insulin enhances membrane transport of glucose, amino acids, and certain ions. It also promotes glycogen storage, formation of triglycerides and synthesis of proteins and nucleic acids. Immunocytochemical investigations have localized insulin in the B cells of pancreatic islets of Langerhans. Deficiency of insulin results in diabetes mellitus, one of the leading causes of morbidity and mortality in the general population. Insulin is also present in tumors of B cell origin such as insulinoma.

包被条件：用5μg/mL抗体包被96孔板，4℃孵育过夜；

检测优化：采用双抗体夹心法，生物素标记二抗+HRP-链霉亲和素系统，提高灵敏度至0.1ng/mL。

IHC-P胰腺组织染色：

抗原修复：采用EDTA缓冲液（pH9.0） 高压修复5分钟，保护胰岛素构象；

阳性定位：胰岛β细胞胞质呈棕黄色颗粒状阳性，α细胞阴性（可与胰高血糖素抗体双标）；

封闭处理：5%驴血清+0.1% Triton X-100室温封闭30分钟，减少非特异性结合。
