产品介绍This gene encodes a member of the interferon regulatory transcription factor (IRF) family. The encoded protein is found in an inactive cytoplasmic form that upon serine/threonine phosphorylation forms a complex with CREBBP. This complex translocates to the nucleus and activates the transcription of interferons alpha and beta, as well as other interferon-induced genes. Alternatively spliced transcript variants encoding multiple isoforms have been observed for this gene. [provided by RefSeq, Nov 2011].

靶点与功能

IRF3是分子量约55kDa的转录因子，静息状态下以单体形式存在于胞质，在病毒感染或TLR3/RLR信号激活后，通过TBK1/IKKε激酶磷酸化C端Ser386位点形成二聚体并入核。其核心功能包括：

抗病毒免疫：激活I型干扰素（IFN-α/β）及干扰素刺激基因（ISGs）表达，是病毒感染早期免疫应答的关键开关；

炎症调控：与NF-κB协同激活促炎因子（如IL-6、TNF-α），维持免疫平衡；

临床关联：在病毒感染性疾病（如流感、COVID-19）、自身免疫病及肿瘤中活性异常，是相关机制研究的重要标志物。

实验操作关键要点

WB检测（病毒感染细胞模型）：

样本处理：HEK293T细胞用VSV病毒感染6小时后，RIPA裂解液（含磷酸酶抑制剂PhosSTOP）冰上裂解30分钟，12000g离心取上清；

电泳条件：10% SDS-PAGE凝胶，恒压80V浓缩胶、120V分离胶，转膜使用0.45μm PVDF膜（恒流300mA 90分钟）；

抗体孵育：一抗1:1000稀释（5%脱脂奶粉+TBST），4℃摇床孵育过夜，HRP二抗1:5000室温孵育1小时，曝光30-60秒（目标条带：非磷酸化约55kDa，磷酸化约57kDa）。

IF核转位实验（TLR3激活模型）：

细胞处理：RAW264.7细胞用poly(I:C)（10μg/mL）刺激4小时，4%多聚甲醛固定15分钟，0.2% Triton X-100透化20分钟；

染色步骤：一抗1:200稀释（含1% BSA的PBS）4℃孵育过夜，Alexa Fluor 488标记二抗（绿色）室温孵育1小时，DAPI（蓝色）染核，激光共聚焦观察IRF3从胞质向核内转移（激活标志）。
