产品介绍This gene encodes a member of the keratin family, the most diverse group of intermediate filaments. This gene product, a type I keratin, is usually found as a heterotetramer with two keratin 5 molecules, a type II keratin. Together they form the cytoskeleton of epithelial cells. Mutations in the genes for these keratins are associated with epidermolysis bullosa simplex. At least one pseudogene has been identified at 17p12-p11. [provided by RefSeq].

靶点与功能

细胞角蛋白14（CK14，Keratin 14）是I型酸性细胞角蛋白（分子量约51kDa），主要表达于复层鳞状上皮的基底细胞及肌上皮细胞，核心研究价值：

上皮分化标记：区分上皮基底细胞与终末分化细胞（如CK10/CK13阳性细胞）；

肿瘤病理诊断：在基底细胞癌、鳞状细胞癌中高表达，用于乳腺癌基底样亚型（三阴性乳腺癌）的鉴别；

疾病关联：CK14基因突变可导致大疱性表皮松解症（EBS），其表达异常与上皮-间质转化（EMT）相关。

实验操作关键要点

IHC-P组织染色（皮肤/乳腺组织）：

抗原修复：采用柠檬酸钠缓冲液（pH6.0） 高压修复3分钟（针对福尔马林固定石蜡切片）；

阳性对照：正常皮肤组织（表皮基底细胞层强阳性）、乳腺肌上皮细胞（胞质阳性）；

显色控制：DAB显色5-10分钟，阳性信号为棕黄色纤维状（与CK5共表达可确认基底样表型）。

WB检测（上皮细胞裂解液）：

样本处理：HaCaT细胞（人永生化角质形成细胞）用含1% SDS的RIPA裂解液冰上裂解30分钟，12000g离心取上清；

电泳条件：10% SDS-PAGE凝胶，恒压80V浓缩胶、120V分离胶，转膜使用0.45μm PVDF膜；

抗体孵育：一抗1:1500稀释（5%脱脂奶粉+TBST），4℃孵育过夜，目标条带约51kDa。
