产品介绍Ki67 antigen is the prototypic cell cycle related nuclear protein, expressed by proliferating cells in all phases of the active cell cycle (G1, S, G2 and M phase). It is absent in resting (G0) cells. Ki67 antibodies are useful in establishing the cell growing fraction in neoplasms (immunohistochemically quantified by determining the number of Ki67 positive cells among the total number of resting cells = Ki67 index). In neoplastic tissues the prognostic value is comparable to the tritiated thymidine labelling index. The correlation between low Ki67 index and histologically low grade tumours is strong. Ki67 is routinely used as a neuronal marker of cell cycling and proliferation.

靶点与功能

Ki67（抗原Ki-67）是一种核抗原，由MKI67基因编码，分子量约359kDa，仅在增殖细胞（G1/S/G2/M期） 中表达，G0期静止细胞不表达。其核心功能包括：

细胞周期标记：作为细胞增殖活性的“金标准”，阳性率（Ki67指数）直接反映组织中处于分裂期的细胞比例；

临床价值：肿瘤Ki67指数与恶性程度正相关（如乳腺癌Ki67>30%提示高侵袭性），是判断预后、指导化疗方案的关键指标；

研究意义：用于评估药物对细胞增殖的抑制效果，或验证细胞周期调控基因的功能。

实验操作关键要点

IHC-P肿瘤组织染色：

抗原修复：采用柠檬酸钠缓冲液（pH6.0） 微波修复20分钟（95℃），冷却后PBS洗涤3次；

阳性定位：细胞核呈棕黄色颗粒状阳性，计数时需避开坏死区，随机选取5个高倍视野（×400）计算阳性细胞百分比（Ki67指数）；

对照设置：必须包含增殖活跃组织（如扁桃体生发中心） 和静止组织（如脑组织） 作为阴阳对照。

WB验证（细胞裂解液）：

样本制备：使用含1mM PMSF和磷酸酶抑制剂的RIPA裂解液冰上裂解30分钟，12000g离心15分钟取上清；

电泳条件：6% SDS-PAGE凝胶（低浓度胶分离大分子量蛋白），恒压60V浓缩胶、80V分离胶，转膜采用0.45μm PVDF膜（250mA恒流2.5小时）；

抗体孵育：兔单抗1:2000稀释（5% BSA+TBST），4℃孵育过夜，HRP二抗（山羊抗兔）1:5000室温孵育1小时。

IF细胞增殖检测：

固定与透化：4%多聚甲醛室温固定15分钟，0.2% Triton X-100透化10分钟；

荧光标记：Alexa Fluor 594偶联二抗（1:1000），DAPI染核，荧光显微镜下计数核阳性细胞比例。
