靶点与功能

Podoplanin（PDPN，又称gp36、T1α）是一种Ⅰ型跨膜糖蛋白，核心功能包括：

淋巴管内皮特异性标记：通过与淋巴管内皮细胞表面的CLEC-2受体结合，调控淋巴管生成（lymphangiogenesis） 和肿瘤淋巴转移（临床数据显示，PDPN高表达肿瘤患者淋巴转移风险增加2.8倍）；

细胞骨架重塑：激活RhoA信号通路促进肌动蛋白聚合，参与血小板聚集、肺泡上皮细胞分化及肾脏足细胞形态维持；

肿瘤微环境调控：在间皮瘤、鳞状细胞癌中高表达，通过诱导肿瘤相关成纤维细胞（CAF）活化促进基质重塑，与患者预后不良显著相关（J Pathol, 2025）。

实验操作关键要点

IHC-P肿瘤淋巴转移研究：

样本处理：福尔马林固定石蜡切片需用EDTA缓冲液（pH9.0） 高压修复15分钟（95℃），增强膜蛋白信号；

阳性定位：正常组织中淋巴管内皮细胞胞膜呈线性阳性，肿瘤组织中可见肿瘤细胞周围新生淋巴管密度增加（需与CD31抗体共染区分血管/淋巴管）；

对照设置：必须包含间皮瘤组织（强阳性对照）和PDPN敲除小鼠组织（阴性对照）。

IF淋巴管可视化：

推荐与LYVE-1抗体共染（淋巴管内皮特异性标记），使用Alexa Fluor 488（绿色，PDPN）和594（红色，LYVE-1），激光共聚焦观察肿瘤边缘淋巴管新生。

WB验证（细胞系）：

样本选择：A549（肺鳞癌，高表达PDPN） vs HUVEC（血管内皮，阴性对照）；

电泳条件：10% SDS-PAGE凝胶，恒压80V浓缩胶、120V分离胶，转膜采用0.45μm PVDF膜（300mA恒流1.5小时）；

抗体孵育：一抗1:1000稀释（5%脱脂奶粉+TBST），4℃孵育过夜，曝光时间60-90秒（糖基化导致条带弥散属正常现象）。
