产品介绍Kallikreins are a subgroup of serine proteases having diverse physiological functions. Growing evidence suggests that many kallikreins are implicated in carcinogenesis and some have potential as novel cancer and other disease biomarkers. This gene is one of the fifteen kallikrein subfamily members located in a cluster on chromosome 19. Its protein product is a protease present in seminal plasma. It is thought to function normally in the liquefaction of seminal coagulum, presumably by hydrolysis of the high molecular mass seminal vesicle protein. Serum level of this protein, called PSA in the clinical setting, is useful in the diagnosis and monitoring of prostatic carcinoma. Alternate splicing of this gene generates several transcript variants encoding different isoforms. [provided by RefSeq, Jul 2008].

靶点与功能

前列腺酸性磷酸酶（PAP，Prostatic Acid Phosphatase）是由前列腺上皮细胞溶酶体分泌的糖蛋白，分子量约100kDa（二聚体形式），等电点4.4-5.5，半衰期1.1-2.6小时。其核心功能包括：

生理作用：参与精液酸性环境维持，促进精子活力；

临床价值：血清PAP水平是前列腺癌骨转移的敏感指标（正常参考值≤2.1μg/L），与PSA联合检测可提高诊断特异性（尤其对PSA“灰区”患者）；

研究意义：通过IHC定位前列腺组织及转移灶中的PAP表达，辅助前列腺癌与膀胱癌、直肠癌的鉴别诊断。

实验操作关键要点

IHC-P组织染色（前列腺癌样本）：

抗原修复：采用柠檬酸钠缓冲液（pH6.0） 微波修复15分钟（95℃），避免EDTA缓冲液影响酶活性；

阳性定位：前列腺上皮细胞胞质呈棕黄色颗粒状阳性，正常组织弥漫分布，癌组织中表达增强且不均；

对照设置：必须包含正常前列腺组织（阳性对照）和膀胱组织（阴性对照，排除交叉反应）。

WB验证（细胞裂解液）：

样本处理：LNCaP细胞（高表达PAP）用含蛋白酶抑制剂（1mM PMSF） 的RIPA裂解液冰上裂解30分钟，12000g离心15分钟取上清；

电泳条件：10% SDS-PAGE凝胶，恒压80V浓缩胶、120V分离胶，转膜使用0.45μm PVDF膜（300mA恒流2小时）；

抗体孵育：一抗1:200稀释（5% BSA+TBST），4℃孵育过夜，HRP二抗（山羊抗小鼠）1:5000室温孵育1小时，曝光时间45-60秒。

ELISA定量检测（血清样本）：

包被条件：抗体用碳酸盐缓冲液（pH9.6）稀释至5μg/mL，4℃包被过夜；

样本处理：血清56℃灭活30分钟，1:50稀释后37℃孵育2小时；

显色控制：TMB底物显色10分钟，2M H₂SO₄终止，450nm读取OD值（标准曲线范围0.1-50ng/mL）。
