产品介绍This gene encodes human lysozyme, whose natural substrate is the bacterial cell wall peptidoglycan (cleaving the beta[1-4]glycosidic linkages between N-acetylmuramic acid and N-acetylglucosamine). Lysozyme is one of the antimicrobial agents found in human milk, and is also present in spleen, lung, kidney, white blood cells, plasma, saliva, and tears. The protein has antibacterial activity against a number of bacterial species. Missense mutations in this gene have been identified in heritable renal amyloidosis. [provided by RefSeq, Oct 2014]

靶点与功能

溶菌酶（Lysozyme）是14kDa糖苷水解酶，通过水解细菌细胞壁肽聚糖的β-1,4糖苷键发挥抗菌作用，广泛存在于眼泪、唾液、乳汁及免疫细胞（如巨噬细胞）中。其核心功能包括：

先天免疫防御：直接裂解革兰氏阳性菌（如金黄色葡萄球菌），协同抗体增强抗菌效果；

炎症调控：通过TLR4信号通路抑制促炎因子释放，在慢性炎症疾病（如慢性阻塞性肺病）中表达异常；

肿瘤微环境：肿瘤相关巨噬细胞分泌溶菌酶可促进肿瘤侵袭，是结直肠癌、肺癌的潜在预后标志物。

实验操作关键要点

WB优化（巨噬细胞样本）：

样本处理：使用含1mM PMSF的RIPA裂解液冰上裂解20分钟，12000g离心15分钟取上清（避免溶菌酶自溶）；

电泳条件：15% SDS-PAGE凝胶（小分子蛋白分离），恒压120V电泳60分钟，转膜采用0.22μm PVDF膜；

抗体孵育：一抗1:1000稀释（5%脱脂奶粉+TBST），4℃孵育过夜，HRP二抗1:5000室温孵育1小时，曝光时间5-10秒。

IHC-P注意事项（肺组织切片）：

抗原修复：采用Tris-EDTA缓冲液（pH9.0） 微波修复15分钟（95℃），增强胞质信号；

阳性对照：推荐正常肺组织（肺泡巨噬细胞胞质呈棕黄色颗粒状阳性）；

封闭处理：5%驴血清+TBST室温封闭30分钟，减少非特异性结合。
