产品介绍The protein encoded by this gene is a member of the RAD51 protein family. RAD51 family members are highly similar to bacterial RecA and Saccharomyces cerevisiae Rad51, and are known to be involved in the homologous recombination and repair of DNA. This protein can interact with the ssDNA-binding protein RPA and RAD52, and it is thought to play roles in homologous pairing and strand transfer of DNA. This protein is also found to interact with BRCA1 and BRCA2, which may be important for the cellular response to DNA damage. BRCA2 is shown to regulate both the intracellular localization and DNA-binding ability of this protein. Loss of these controls following BRCA2 inactivation may be a key event leading to genomic instability and tumorigenesis. Two alternatively spliced transcript variants of this gene, which encode distinct proteins, have been reported. Transcript variants utilizing alternative polyA signals exist.

RALA（Ras-like protein A）是RAS超家族小GTP酶成员，通过GTP结合/水解的分子开关机制，调控细胞骨架重塑、膜泡运输及上皮-间质转化（EMT）。其核心功能包括：

肿瘤进展：激活下游效应因子RALBP1促进肿瘤细胞侵袭，在胰腺癌中与转移风险正相关（临床样本数据显示高表达者5年生存率降低42%）；

细胞极性维持：与Exocyst复合体结合参与细胞顶-基底极性建立，缺失可导致上皮细胞紧密连接破坏；

病毒复制调控：作为HIV-1病毒出芽的关键宿主因子，通过与Gag蛋白互作促进病毒颗粒释放。

实验操作关键要点

WB优化（肿瘤细胞样本）：

样本处理：使用含GDP解离抑制剂（如RhoGDI） 的RIPA裂解液冰上裂解30分钟，12000g离心15分钟取上清；推荐使用PANC-1胰腺癌细胞系作为阳性对照（RALA高表达）；

电泳条件：12% SDS-PAGE凝胶，恒压80V浓缩胶、120V分离胶，电泳时间80分钟确保24kDa条带分离；

抗体孵育：一抗推荐1:2000稀释（5%脱脂奶粉+TBST），4℃摇床孵育过夜，HRP标记二抗（山羊抗兔）1:5000室温孵育1小时，曝光时间30-60秒。

IF双标实验（细胞定位研究）：

推荐与β-catenin抗体共染（上皮细胞极性标志物），使用Alexa Fluor 488（绿色，RALA主要定位于细胞膜/胞质）和594（红色，β-catenin定位于细胞连接），激光共聚焦观察EMT过程中的共定位变化。

IP实验注意事项：

磁珠选择：使用Protein A/G beads，每500μg总蛋白加入5μg抗体，4℃旋转孵育4小时；

洗脱条件：0.1M甘氨酸-HCl（pH2.8）洗脱后立即用1M Tris-HCl（pH8.0）中和，避免抗原抗体复合物解离不完全。
