Key regulator of LPAR1 signaling and competes with ADRBK1 for binding to LPAR1 thus affecting the signaling properties of the receptor. Required for anchorage-independent proliferation of transformed cells.

靶点与功能

S6核糖体蛋白（RPS6，40S ribosomal protein S6）是40S核糖体亚基的组成部分，通过磷酸化修饰（如Ser235/236、Ser240/244位点）参与mRNA翻译起始调控，是mTOR信号通路的关键下游效应分子。其磷酸化激活可促进核糖体生物合成和细胞周期进展，与细胞增殖、肿瘤发生（如乳腺癌、结直肠癌） 及代谢疾病密切相关，是研究细胞生长调控的重要标志物。

实验操作关键要点

WB优化（肿瘤细胞样本）：

样本处理：使用含磷酸酶抑制剂（如PhosSTOP） 的RIPA裂解液冰上裂解30分钟，12000g离心15分钟取上清；推荐使用HeLa细胞系（RPS6高表达）作为阳性对照；

电泳条件：10% SDS-PAGE凝胶，恒压80V浓缩胶、120V分离胶，电泳时间90分钟确保29kDa条带分离；

抗体孵育：一抗推荐1:1000稀释（5%脱脂奶粉+TBST），4℃摇床孵育过夜，HRP标记二抗（山羊抗小鼠）1:5000室温孵育1小时，曝光时间30-60秒。

IHC-P注意事项（结肠癌组织）：

抗原修复：采用柠檬酸钠缓冲液（pH6.0） 微波修复20分钟（95℃），增强核周/胞质信号；

阳性定位：肿瘤细胞胞质呈棕黄色颗粒状阳性，正常组织中表达较低；

封闭处理：5%羊血清+TBST室温封闭30分钟，减少非特异性结合。

IF双标实验（mTOR通路研究）：

推荐与磷酸化S6K1（p-S6K1，Thr389） 共染，使用Alexa Fluor 488（绿色，标记RPS6）和594（红色，标记p-S6K1），激光共聚焦观察二者共定位（激活的mTOR信号通路）。
