产品介绍Ribosomes, the organelles that catalyze protein synthesis, consist of a small 40S subunit and a large 60S subunit. Together these subunits are composed of 4 RNA species and approximately 80 structurally distinct proteins. This gene encodes a cytoplasmic ribosomal protein that is a component of the 40S subunit. The protein belongs to the S6E family of ribosomal proteins. It is the major substrate of protein kinases in the ribosome, with subsets of five C-terminal serine residues phosphorylated by different protein kinases. Phosphorylation is induced by a wide range of stimuli, including growth factors, tumor-promoting agents, and mitogens. Dephosphorylation occurs at growth arrest. The protein may contribute to the control of cell growth and proliferation through the selective translation of particular classes of mRNA. As is typical for genes encoding ribosomal proteins, there are multiple processed pseudogenes of this gene dispersed through the genome. [provided by RefSeq, Jul 2008]

靶点与功能

S100B（S100 calcium-binding protein B）是EF-hand钙结合蛋白家族成员，主要由星形胶质细胞、少突胶质细胞及黑色素瘤细胞分泌，通过与钙结合调控细胞增殖、神经突延伸及炎症反应。其核心功能包括：

神经损伤标志物：脑卒中后血液/脑脊液S100B水平升高，可作为神经损伤程度的量化指标（临床阈值>0.5 ng/mL提示中度脑损伤）；

肿瘤进展调控：在黑色素瘤中通过激活RAGE/NF-κB通路促进转移，与患者不良预后显著相关（Ⅲ期黑色素瘤中高表达者5年生存率降低32%）；

神经修复双重作用：纳摩尔浓度促进神经再生，微摩尔浓度诱导神经元凋亡。

实验操作关键要点

WB优化（脑组织/黑色素瘤样本）：

样本处理：使用含1mM CaCl₂的RIPA裂解液（维持S100B活性构象）冰上裂解25分钟，14000g离心20分钟取上清；

电泳条件：15% SDS-PAGE凝胶，恒压80V浓缩胶、120V分离胶，电泳时间50分钟，确保10kDa单体与20kDa二聚体分离；

抗体孵育：一抗推荐1:1000稀释（5% BSA+TBST），4℃摇床孵育过夜，HRP二抗（山羊抗兔）1:5000室温孵育1小时，曝光时间8-15秒。

IHC-P注意事项（脑胶质瘤组织）：

抗原修复：采用柠檬酸钠缓冲液（pH6.0） 微波修复15分钟（95℃），避免EDTA螯合钙离子；

阳性定位：星形胶质细胞胞质呈棕黄色颗粒状阳性，瘤周组织表达增强；

封闭处理：5%驴血清+0.3% Triton X-100室温封闭45分钟，减少非特异性结合。

IF双标实验（神经炎症研究）：

推荐与GFAP抗体（星形胶质细胞标志物） 共染，使用Alexa Fluor 488（绿色，S100B）和594（红色，GFAP），激光共聚焦观察脑损伤区域胶质细胞活化。
