产品介绍S100 beta is a member of the S100 family of proteins containing 2 EF-hand calcium binding motifs. S100 proteins are localized in the cytoplasm and/or nucleus of a wide range of cells, and involved in the regulation of a number of cellular processes such as cell cycle progression and differentiation. S100 genes include at least 13 members which are located as a cluster on chromosome 1q21; however, this gene is located at 21q22.3. This protein may function in neurite extension, proliferation of melanoma cells, stimulation of Ca2+ fluxes, inhibition of PKC mediated phosphorylation, astrocytosis and axonal proliferation, and inhibition of microtubule assembly. Chromosomal rearrangements and altered expression of this gene have been implicated in several neurological, neoplastic, and other types of diseases, including Alzheimer's disease, Down's syndrome, epilepsy, amyotrophic lateral sclerosis, melanoma, and type I diabetes.

靶点与功能

S100A1（S100 calcium-binding protein A1）是EF-hand结构域钙结合蛋白，主要表达于心肌、骨骼肌及神经系统，通过与钙结合后构象变化，调控肌浆网钙释放通道（RyR2）活性、细胞能量代谢及细胞骨架动态。其表达异常与心力衰竭（心肌S100A1水平降低）、阿尔茨海默病（脑内聚集） 及乳腺癌（高表达促转移）密切相关，是心血管疾病和肿瘤研究的重要标志物。

实验操作关键要点

WB优化（心肌组织样本）：

样本处理：使用含1mM CaCl₂的RIPA裂解液（维持S100A1钙结合活性）冰上裂解20分钟，12000g离心15分钟取上清；

电泳条件：15% SDS-PAGE凝胶（因分子量小），恒压80V浓缩胶、120V分离胶，电泳时间60分钟；

抗体孵育：一抗推荐1:1000稀释（5%脱脂奶粉+TBST），4℃摇床孵育过夜，HRP标记二抗（山羊抗兔）1:5000室温孵育1小时，曝光时间5-10秒（避免信号过曝）。

IHC-P注意事项（心脏组织切片）：

抗原修复：采用柠檬酸钠缓冲液（pH6.0） 微波修复15分钟（95℃），避免EDTA缓冲液螯合钙离子影响抗原表位；

阳性对照：推荐使用正常心肌组织（心肌细胞胞质呈棕黄色弥漫性阳性）；

封闭处理：5%羊血清+TBST室温封闭30分钟，减少非特异性结合。

ELISA检测要点（血清样本）：

包被条件：5μg/mL抗体包被96孔板，4℃孵育过夜，PBST洗涤3次；

样本稀释：血清用含1% BSA的PBS按1:100稀释，37℃孵育2小时；

显色控制：TMB底物显色10分钟，2M H₂SO₄终止反应，450nm波长读取OD值。
