S100 proteins are localized in the cytoplasm and/or nucleus of a wide range of cells, and involved in the regulation of a number of cellular processes such as cell cycle progression and differentiation. 靶点与功能

FSP-1（Fibroblast Specific Protein 1，S100A4）是S100钙结合蛋白家族成员，通过调控细胞骨架重塑和细胞外基质（ECM）降解参与成纤维细胞活化、上皮-间质转化（EMT） 过程。其高表达与肝纤维化、肺纤维化及肿瘤侵袭转移密切相关，是活化成纤维细胞（肌成纤维细胞）和肿瘤相关成纤维细胞（CAF）的特异性标志物，也是评估纤维化程度和肿瘤微环境的关键指标。

实验操作关键要点

IHC-P优化（纤维化组织样本）：

样本处理：福尔马林固定石蜡切片需用柠檬酸钠缓冲液（pH6.0） 微波修复20分钟（95℃），增强胞质信号；

抗体孵育：一抗推荐1:100稀释（5%羊血清+TBST），4℃湿盒孵育过夜，HRP标记二抗（山羊抗小鼠）1:500室温孵育30分钟；

阳性对照：推荐使用肝纤维化模型小鼠肝组织（汇管区成纤维细胞胞质呈棕黄色阳性）。

WB检测要点（成纤维细胞样本）：

样本处理：使用含蛋白酶抑制剂（1mM PMSF） 的RIPA裂解液冰上裂解30分钟，12000g离心15分钟取上清；

电泳条件：15% SDS-PAGE凝胶（因分子量小），恒压80V浓缩胶、120V分离胶，确保11kDa主条带分离；

转膜条件：PVDF膜，恒流300mA转膜60分钟（冰浴），避免小分子蛋白穿透膜。

IF双标实验（肿瘤微环境研究）：

推荐与α-SMA抗体共染（肌成纤维细胞标志物），使用Alexa Fluor 488（绿色，标记FSP-1）和594（红色，标记α-SMA），激光共聚焦观察CAF的双阳性表达。
