产品介绍The major constituents of mucus, the viscous secretion that covers epithelial surfaces such as those in the trachea, colon, and cervix, are highly glycosylated proteins called mucins. These glycoproteins play important roles in the protection of the epithelial cells and have been implicated in epithelial renewal and differentiation. This gene encodes an integral membrane glycoprotein found on the cell surface, although secreted isoforms may exist. At least two dozen transcript variants of this gene have been found, although for many of them the full-length transcript has not been determined or they are found only in tumor tissues. This gene contains a region in the coding sequence which has a variable number (>100) of 48 nt tandem repeats. [provided by RefSeq].

靶点与功能

胃粘液素主要包括MUC5AC（胃体/胃底表面黏液细胞） 和MUC6（胃腺颈黏液细胞/幽门腺），核心功能与研究价值：

胃黏膜屏障核心：MUC5AC形成凝胶状黏液层，隔离胃酸（pH 1-2）与上皮细胞；MUC6分泌碳酸氢盐，维持黏液层pH梯度（近上皮侧pH 7.0），二者共同构成“黏液-碳酸氢盐屏障”（破坏后易引发幽门螺杆菌定植）；

疾病标志物：胃癌中MUC5AC表达下调（肠型胃癌阳性率仅15%），而MUC6在胃底腺息肉中高表达（与β-catenin通路激活相关）；

临床转化：血清MUC5AC水平可作为胃溃疡愈合的监测指标（愈合期较急性期升高2.3倍）。

实验操作关键要点

IHC-P胃组织染色：

抗原修复：柠檬酸盐缓冲液（pH6.0）高压修复10分钟（石蜡切片），MUC5AC在胃小凹表面上皮细胞胞质呈棕黄色，MUC6在胃腺颈部细胞呈强阳性；

阳性对照：正常胃体组织（MUC5AC+/MUC6+），阴性对照：十二指肠组织（MUC5AC-）；

病理特征：幽门螺杆菌感染样本中MUC5AC阳性面积较正常组减少28%（ImageJ定量）。

WB检测（胃黏膜样本）：

样本处理：新鲜胃黏膜用含蛋白酶抑制剂（1mM PMSF） 和1% Triton X-100的RIPA裂解液冰上裂解30分钟，12000g离心取上清；

电泳条件：6% SDS-PAGE凝胶，恒压60V浓缩胶、80V分离胶，转膜使用0.45μm PVDF膜（300mA恒流2.5小时）；

抗体孵育：一抗1:1000稀释（5%脱脂奶粉+TBST），4℃孵育过夜，HRP二抗（山羊抗兔）1:5000室温孵育1小时。

IF双标实验：

MUC5AC（绿色）与Ki-67（红色） 共染，胃小凹底部增殖细胞（Ki-67+）同时表达MUC5AC，提示干细胞分化方向；

固定条件：4%多聚甲醛固定15分钟，0.2% Triton X-100透化10分钟，5% BSA封闭30分钟。
