产品介绍The protein encoded by this gene is a member of the S100 family of proteins containing 2 EF-hand calcium-binding motifs. S100 proteins are localized in the cytoplasm and/or nucleus of a wide range of cells, and involved in the regulation of a number of cellular processes such as cell cycle progression and differentiation. S100 genes include at least 13 members which are located as a cluster on chromosome 1q21. This protein may function in motility, invasion, and tubulin polymerization. Chromosomal rearrangements and altered expression of this gene have been implicated in tumor metastasis. Multiple alternatively spliced variants, encoding the same protein, have been identified. 靶点与功能

S100A6（Calcyclin）是S100家族钙结合蛋白成员，通过EF-hand结构域结合Ca²⁺后发生构象变化，调控细胞增殖、侵袭及信号转导。其表达异常与乳腺癌、结直肠癌、胰腺癌等肿瘤的恶性进展密切相关，可通过与靶蛋白（如Annexin II、p53）相互作用促进上皮-间质转化（EMT），同时参与炎症反应中的中性粒细胞募集，是肿瘤诊断和预后评估的潜在标志物。

实验操作关键要点

WB优化（肿瘤细胞样本）：

样本处理：使用含蛋白酶抑制剂（1mM PMSF） 的RIPA裂解液冰上裂解30分钟，12000g离心15分钟取上清；若检测钙结合活性，需在裂解液中添加1mM CaCl₂（避免S100A6构象失活）；

电泳条件：15% SDS-PAGE凝胶（因分子量小），恒压80V浓缩胶、120V分离胶，确保10kDa单体与20kDa二聚体条带分离；

抗体孵育：一抗推荐1:1000稀释（5% BSA+TBST），4℃摇床孵育过夜，HRP标记二抗（山羊抗兔）1:5000室温孵育1小时，曝光时间控制在15-30秒避免信号过强。

IHC-P注意事项（乳腺癌组织切片）：

抗原修复：采用柠檬酸钠缓冲液（pH6.0） 微波修复15分钟（95℃），增强胞质信号；

阳性对照：推荐使用乳腺浸润性导管癌组织（癌细胞胞质呈棕黄色颗粒状阳性，间质细胞弱阳性）；

封闭处理：5%驴血清+0.3% Triton X-100室温封闭45分钟，降低非特异性结合。

IF双标实验（钙信号共定位研究）：

推荐与Annexin II抗体共染（膜定位蛋白），使用Alexa Fluor 488（绿色，标记S100A6）和594（红色，标记Annexin II），激光共聚焦观察二者在细胞边缘的共定位（促进肿瘤细胞迁移的分子复合物）。
