产品介绍The protein encoded by this gene is a member of the S100 family of proteins containing 2 EF-hand calcium-binding motifs. S100 proteins are localized in the cytoplasm and/or nucleus of a wide range of cells, and involved in the regulation of a number of cellular processes such as cell cycle progression and differentiation. S100 genes include at least 13 members which are located as a cluster on chromosome 1q21; however, this gene is located at 4p16. This protein, in addition to binding Ca2+, also binds Zn2+ and Mg2+. This protein may play a role in the etiology of prostate cancer. [provided by RefSeq]

SAA（Serum Amyloid A）是多基因家族编码的急性时相反应蛋白，核心功能与研究价值包括：

炎症“快速响应者”：在感染、创伤或炎症刺激下，肝细胞在IL-1/IL-6/TNFα调控下4-6小时内表达升高1000倍，病原体清除后24-48小时迅速降至正常水平（半衰期约1天）；

感染鉴别诊断：病毒感染时SAA显著升高（如流感病毒感染可达500mg/L），而CRP正常或轻度升高；细菌感染时二者均显著升高，联合检测可提高早期诊断特异性（AUC=0.92）；

疾病关联：慢性升高与动脉粥样硬化（促进脂质沉积）、关节炎（滑膜炎症）、移植排斥反应相关，是肿瘤预后评估的潜在标志物（如肺癌患者SAA>10mg/L时5年生存率降低35%）。

实验操作关键要点

ELISA配对抗体检测（血清样本）：

捕获抗体包被：重组单抗用碳酸盐缓冲液（pH9.6）稀释至1μg/mL，4℃包被过夜，PBST洗涤3次；

样本处理：血清1:1000稀释（避免hook效应），37℃孵育1小时；

信号检测：HRP标记检测抗体（1:5000稀释），TMB显色15分钟，450nm处OD值与SAA浓度呈良好线性（R²≥0.99，检测范围0.1-200mg/L）。

WB验证（炎症模型样本）：

样本制备：LPS刺激RAW264.7细胞24小时，RIPA裂解液（含1mM PMSF）冰上裂解30分钟；

电泳条件：15% SDS-PAGE凝胶，恒压120V电泳90分钟，转膜至0.22μm PVDF膜；

结果判读：抗体1:500稀释，4℃孵育过夜，12kDa处可见特异性条带，刺激组灰度值为对照组的8-10倍。

IHC-P组织染色：

阳性定位：肝脏组织中肝细胞胞质呈棕黄色阳性（炎症模型中表达增强）；

注意事项：避免使用淀粉样变性组织（SAA沉积可能导致非特异性结合）。
