产品介绍This gene encodes a member of the serum amyloid A family of apolipoproteins. The encoded protein is a major acute phase protein that is highly expressed in response to inflammation and tissue injury. This protein also plays an important role in HDL metabolism and cholesterol homeostasis. High levels of this protein are associated with chronic inflammatory diseases including atherosclerosis, rheumatoid arthritis, Alzheimer's disease and Crohn's disease. This protein may also be a potential biomarker for certain tumors. Alternate splicing results in multiple transcript variants that encode the same protein. A pseudogene of this gene is found on chromosome 11.[provided by RefSeq, Jun 2012]

SLC44A1（Solute Carrier Family 44 Member 1）又称CD92或胆碱转运蛋白样蛋白1（CHTL1），是一种Na⁺非依赖性胆碱转运蛋白，主要参与细胞膜磷脂合成和神经髓鞘生成，在神经胶质细胞分化、肿瘤细胞代谢中发挥调控作用。其表达异常与多发性硬化、肝癌转移等疾病相关，是研究胆碱代谢通路的重要分子标志物。

实验操作关键要点

WB优化（神经胶质细胞样本）：

样本处理：使用含蛋白酶抑制剂（1mM PMSF） 的RIPA裂解液冰上裂解30分钟，12000g离心15分钟取上清；推荐使用C6胶质瘤细胞系作为阳性对照（SLC44A1高表达）；

电泳条件：10% SDS-PAGE凝胶，恒压80V浓缩胶、120V分离胶，确保73kDa主条带与杂带分离；

抗体孵育：一抗推荐1:1500稀释（5% BSA+TBST），4℃摇床孵育过夜，HRP标记二抗（山羊抗兔）1:5000室温孵育1小时。

IHC-P注意事项（脑组织样本）：

抗原修复：采用柠檬酸钠缓冲液（pH6.0） 微波修复20分钟（95℃），增强膜蛋白信号；

阳性定位：正常脑组织中少突胶质细胞胞膜呈棕黄色阳性，肿瘤组织中可见细胞膜/胞质弥漫性表达；

封闭处理：5%驴血清+0.3% Triton X-100室温封闭45分钟，降低非特异性结合。

IF双标实验（细胞定位研究）：

推荐与MBP（髓鞘碱性蛋白） 共染，使用Alexa Fluor 488（绿色，标记SLC44A1）和594（红色，标记MBP），激光共聚焦观察少突胶质细胞共定位。
