产品介绍The protein encoded by this gene binds copper and zinc ions and is one of two isozymes responsible for destroying free superoxide radicals in the body. The encoded isozyme is a soluble cytoplasmic protein, acting as a homodimer to convert naturally-occuring but harmful superoxide radicals to molecular oxygen and hydrogen peroxide. The other isozyme is a mitochondrial protein. Mutations in this gene have been implicated as causes of familial amyotrophic lateral sclerosis. 靶点与功能

钠钾ATP酶α1（Na⁺/K⁺-ATPase α1亚基）是细胞膜上的跨膜蛋白，通过水解ATP逆浓度梯度转运Na⁺（胞外）和K⁺（胞内），维持细胞静息电位、渗透压平衡及神经肌肉兴奋性。其表达异常与高血压、心力衰竭、肾小管酸中毒等密切相关，是强心苷类药物（如地高辛）的作用靶点，也是评估细胞膜完整性和细胞活力的重要标志物。

实验操作关键要点

WB优化（细胞膜蛋白提取）：

样本处理：使用膜蛋白提取试剂盒分离细胞膜组分，避免胞质蛋白干扰；裂解液需添加蛋白酶抑制剂（PMSF+EDTA） 和磷酸酶抑制剂，冰上操作30分钟；

电泳条件：7.5% SDS-PAGE凝胶（因分子量较大），恒压80V浓缩胶、120V分离胶，电泳时间90分钟确保条带分离；

抗体孵育：一抗推荐1:2000稀释（5%脱脂奶粉+TBST），4℃摇床孵育过夜，HRP标记二抗（山羊抗兔）1:5000室温孵育1小时，曝光时间控制在60-90秒。

IHC-P注意事项（肾脏/心肌组织）：

抗原修复：采用EDTA缓冲液（pH9.0） 高压修复10分钟（针对石蜡切片），增强细胞膜信号；

阳性对照：推荐使用大鼠肾脏皮质组织（近曲小管上皮细胞膜呈棕黄色强阳性）；

封闭处理：5%牛血清白蛋白（BSA）室温封闭60分钟，减少非特异性结合。

IF双标实验（细胞极性研究）：

推荐与紧密连接蛋白ZO-1共染，使用Alexa Fluor 488（绿色，标记NKAα1，基底侧膜定位）和594（红色，标记ZO-1，顶侧膜定位），激光共聚焦观察上皮细胞极性分布。
