产品介绍The protein encoded by this gene belongs to the family of P-type cation transport ATPases, and to the subfamily of Na+/K+-ATPases. Na+/K+ -ATPase is an integral membrane protein responsible for establishing and maintaining the electrochemical gradients of Na and K ions across the plasma membrane. These gradients are essential for osmoregulation, for sodium-coupled transport of a variety of organic and inorganic molecules, and for electrical excitability of nerve and muscle. This enzyme is composed of two subunits, a large catalytic subunit (alpha) and a smaller glycoprotein subunit (beta). The catalytic subunit of Na+/K+ -ATPase is encoded by multiple genes. This gene encodes an alpha 1 subunit. Multiple transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, May2009].

靶点与功能

转录因子SOX10（SRY-related HMG-box gene 10）是SOX家族C亚族成员，通过结合靶基因启动子的HMG结构域调控转录，在神经嵴细胞分化、周围神经系统发育及黑色素细胞生成中起核心作用。其功能异常与神经嵴来源肿瘤（如黑色素瘤、神经鞘瘤、雪旺细胞瘤）密切相关，是这类肿瘤病理诊断的特异性标志物，也参与肌上皮肿瘤的鉴别诊断。

实验操作关键要点

IHC-P优化（黑色素瘤病理诊断）：

样本处理：福尔马林固定石蜡包埋组织需用pH9.0 EDTA缓冲液热诱导抗原修复（95℃，20分钟），充分暴露核内抗原表位；

抗体孵育：一抗推荐1:200稀释（5%驴血清+TBST），4℃湿盒孵育过夜，HRP标记二抗（山羊抗兔）1:500室温孵育30分钟；

显色控制：DAB显色时间3-5分钟，阳性信号定位于肿瘤细胞核（呈棕黄色颗粒状），阴性对照（正常皮肤组织）仅表皮基底层少量黑色素细胞阳性。

WB检测要点（神经嵴细胞样本）：

样本处理：使用含蛋白酶抑制剂（1mM PMSF） 的RIPA裂解液冰上裂解30分钟，12000g离心15分钟取上清；

电泳条件：10% SDS-PAGE凝胶，恒压80V浓缩胶、120V分离胶，确保47kDa主条带与杂带分离；

阳性对照：推荐使用SK-MEL-28黑色素瘤细胞裂解液（47kDa条带清晰可见）。

IF双标实验（细胞定位研究）：

推荐与S100蛋白共染，使用Alexa Fluor 488（绿色，标记SOX10，核定位）和594（红色，标记S100，胞质定位），激光共聚焦观察神经嵴细胞共表达模式。
