产品介绍Mucin is a high M.W. (>1,000 kDa) glycoprotein, expressed by mucus cells of the gastric epithelium and by goblet cells of the fetal, precancerous and cancerous colon, but not by those of the normal colon. It also appears in other epithelial tissues, which are embryologically derived from the foregut (epigastric and bronchial epithelium) and in Müller ducts (mucus cells of the endocervix and urethral epithelium near the prostatic utriculus). The expression of MUC5AC has been frequently observed in gastric carcinomas and may be used as a marker of gastric differentiation.

靶点与功能

MUC6是凝胶形成型粘蛋白家族成员（分子量>500kDa），核心功能与研究价值包括：

胃黏膜屏障基石：由胃体腺颈黏液细胞和幽门腺细胞分泌，在胃黏膜表面形成黏液-碳酸氢盐屏障，物理阻隔胃酸（pH1-2）和胃蛋白酶对上皮细胞的侵蚀；

疾病标志物：慢性萎缩性胃炎时MUC6表达区域缩小（幽门腺化生），胃癌前病变（肠上皮化生）中MUC6⁺细胞比例降低>50%，是病理诊断的重要指标；

抗菌防御作用：通过O-糖链结合幽门螺杆菌（Hp）的黏附素，抑制其定植（MUC6缺失小鼠Hp感染率提高3倍）。

实验操作关键要点

IHC-P胃组织染色：

抗原修复：柠檬酸盐缓冲液（pH6.0）微波修复20分钟（石蜡切片），幽门腺区域呈棕黄色强阳性（胃体腺颈黏液细胞弱阳性）；

病理特征：正常胃黏膜MUC6表达于幽门腺全层，慢性萎缩性胃炎时阳性区域向胃体扩展（假幽门腺化生）；

去糖基化处理：若使用识别核心肽段的抗体（如ab213447），需用0.5U/μL PNGase F 37℃处理切片1小时，暴露抗原表位。

WB检测（胃黏膜组织）：

样本处理：新鲜胃黏膜用含蛋白酶抑制剂（1mM PMSF） 的RIPA裂解液（添加1% NP-40）冰上裂解30分钟，12000g离心取上清；

电泳条件：4-15%梯度SDS-PAGE凝胶，恒压80V浓缩胶、100V分离胶，电泳150分钟（因分子量极大）；

结果特征：天然MUC6在WB中呈>250kDa的弥散条带，去糖基化后条带迁移至180kDa左右。

ELISA检测（胃液样本）：

包被抗体用碳酸盐缓冲液（pH9.6）稀释至5μg/mL，4℃包被过夜；

胃液样本需经10kDa超滤管离心（3000g，30分钟）去除小分子杂质，1:50稀释后检测；

临床意义：胃溃疡患者胃液MUC6浓度较健康人降低42%（ELISA定量）。
