产品介绍MUC1 is a large cell surface mucin glycoprotein expressed by most glandular and ductal epithelial cells and some hematopoietic cell lineages. It is expressed on most secretory epithelium, including mammary gland and some hematopoietic cells. It is expressed abundantly in lactating mammary glands and overexpressed abundantly in >90% breast carcinomas and metastases. Transgenic MUC1 has been shown to associate with all four cebB receptors and localize with erbB1 (EGFR) in lactating glands. The MUC1 gene contains seven exons and produces several different alternatively spliced variants. The major expressed form of MUC1 uses all seven exons and is a type 1 transmembrane protein with a large extracellular tandem repeat domain. The tandem repeat domain is highly O glycosylated and alterations in glycosylation have been shown in epithelial cancer cells.

靶点与功能

MYBL2（B-Myb）是MYB家族转录因子，分子量约77-79kDa，核心功能与研究价值包括：

细胞周期调控：通过激活Cyclin A、CDK2等靶基因，调控G1/S期转换（MYBL2缺失导致细胞周期停滞于G1期）；

肿瘤关联：在乳腺癌（ER⁺亚型）、肺癌、淋巴瘤中高表达，与肿瘤分级、淋巴结转移正相关（MYBL2高表达患者5年生存率降低40%）；

胚胎发育：参与造血干细胞分化和神经发生，敲除小鼠因胎盘发育缺陷导致胚胎致死。

实验操作关键要点

WB检测（细胞周期同步化样本）：

样本处理：HeLa细胞经胸腺嘧啶核苷双阻断法同步化至G1/S期，RIPA裂解液（含1mM PMSF+PhosSTOP）冰上裂解30分钟；

结果特征：总MYBL2抗体在77kDa处显带，磷酸化抗体（Ser577）在G2/M期出现特异性条带（与CDK1磷酸化活性相关）；

内参选择：建议用Cyclin A作为阳性对照（二者表达趋势一致）。

IHC-P肿瘤组织染色：

抗原修复：柠檬酸盐缓冲液（pH6.0）微波修复20分钟（石蜡切片），肿瘤细胞核呈棕黄色阳性；

阳性定位：乳腺癌组织中ER⁺细胞核阳性率显著高于ER⁻细胞（MYBL2与ER信号通路协同作用）；

对照设置：正常乳腺组织（弱阳性）和MYBL2敲除细胞系（阴性对照）。

IF核定位实验：

与Ki-67抗体共染，MYBL2（红色，核定位）与Ki-67（绿色，增殖标志物）共表达率>80%提示高增殖活性细胞群。
