The encoded protein may act as a transcriptional activator after forming a protein complex with other proteins. This protein acts as a nucleocytoplasmic shuttle protein and is important for neural crest and peripheral nervous system development. Oct4：通过结合靶基因启动子的八聚体序列，调控胚胎发育早期细胞命运决定，是干细胞多能性的标志性标志物；

Nanog：维持干细胞未分化状态，抑制分化相关基因表达，与Oct4/Sox2协同构成多能性核心调控轴；

Sox2：参与胚胎神经外胚层发育，与Oct4共同激活多能性基因（如Utf1、Rex1）的转录。

实验操作关键要点

WB优化（胚胎干细胞样本）：

样本处理：使用含 磷酸酶抑制剂（PhosSTOP） 的RIPA裂解液冰上裂解30分钟，12000g离心15分钟取上清，避免转录因子降解；

电泳条件：10% SDS-PAGE凝胶，恒压80V浓缩胶、120V分离胶，确保43kDa主条带清晰；

抗体孵育：一抗推荐1:2000稀释（5% BSA+TBST），4℃摇床孵育过夜，HRP标记二抗（山羊抗兔）1:5000室温孵育1小时。

IHC-P注意事项（胚胎组织切片）：

抗原修复：采用 柠檬酸钠缓冲液（pH6.0） 微波修复20分钟（95℃），增强细胞核信号；

阳性对照：推荐使用 人胚胎干细胞（hESC）爬片（细胞核呈棕黄色阳性）；

封闭处理：5%驴血清+0.3% Triton X-100室温封闭45分钟，降低非特异性背景。

ChIP实验要点（多能性基因调控研究）：

染色质制备：使用 甲醛（1%终浓度） 室温交联10分钟，甘氨酸终止后超声破碎（片段长度200-500bp）；

抗体孵育：5μg抗体/1×10⁶细胞量的染色质，4℃旋转孵育过夜，Protein A/G磁珠富集DNA-蛋白复合物。
