产品介绍This intronless gene encodes a member of the SRY-related HMG-box (SOX) family of transcription factors involved in the regulation of embryonic development and in the determination of cell fate. The product of this gene is required for stem-cell maintenance in the central nervous system, and also regulates gene expression in the stomach. Mutations in this gene have been associated with optic nerve hypoplasia and with syndromic microphthalmia, a severe form of structural eye malformation. This gene lies within an intron of another gene called SOX2 overlapping transcript (SOX2OT). [provided by RefSeq, Jul 2008].

靶点与功能

P62（Sequestosome 1，SQSTM1）是一种多功能支架蛋白，通过其UBA结构域结合泛素化蛋白，介导异常蛋白通过自噬-溶酶体途径降解，同时参与NF-κB信号激活、Nrf2抗氧化通路调控及细胞凋亡抑制。其功能异常与癌症（如肝癌、肺癌）、神经退行性疾病（如帕金森病） 及代谢综合征直接相关，是自噬 flux 检测和疾病机制研究的关键标志物。

实验操作关键要点

WB优化（自噬 flux 检测）：

样本处理：细胞需用含蛋白酶抑制剂（PMSF+抑肽酶） 的RIPA裂解液冰上裂解30分钟，14000g离心20分钟取上清；若检测自噬流，需同步设置氯喹（CQ，50μM）处理组作为阳性对照（抑制溶酶体降解，P62累积）；

电泳条件：10% SDS-PAGE凝胶，恒压80V浓缩胶、120V分离胶，确保62kDa主条带与修饰型条带分离；

抗体孵育：一抗推荐1:2000稀释（5%脱脂奶粉+TBST），4℃摇床孵育过夜，HRP标记二抗（山羊抗兔）1:5000室温孵育1小时。

IHC-P注意事项（肿瘤组织样本）：

抗原修复：采用EDTA缓冲液（pH9.0） 高压修复5分钟（针对福尔马林固定石蜡切片），增强胞质信号；

阳性对照：推荐使用肝癌组织切片（肿瘤细胞胞质呈棕黄色颗粒状阳性，伴核周聚集）；

封闭处理：5%驴血清+0.3% Triton X-100室温封闭45分钟，配合3% H₂O₂阻断内源性过氧化物酶（15分钟）。

IF双标实验（自噬共定位研究）：

推荐与LC3B抗体共染（自噬小体标志物），使用Alexa Fluor 488（绿色，标记P62）和594（红色，标记LC3B），激光共聚焦观察二者共定位斑点（自噬降解复合物）。
