产品介绍This gene encodes a multifunctional protein that binds ubiquitin and regulates activation of the nuclear factor kappa-B (NF-kB) signaling pathway. The protein functions as a scaffolding/adaptor protein in concert with TNF receptor-associated factor 6 to mediate activation of NF-kB in response to upstream signals. Alternatively spliced transcript variants encoding either the same or different isoforms have been identified for this gene. Mutations in this gene result in sporadic and familial Paget disease of bone. [provided by RefSeq, Mar 2009]

靶点与功能

STAT5（Signal Transducer and Activator of Transcription 5）是STAT家族成员，包括STAT5A和STAT5B两个高度同源亚型，通过细胞因子（如IL-2、IL-3、EPO）激活的JAK激酶磷酸化后入核，调控细胞增殖、分化及抗凋亡基因（如Bcl-2、Cyclin D1）的转录。其持续激活与慢性粒细胞白血病（CML）、乳腺浸润性导管癌、前列腺癌等密切相关，是肿瘤靶向治疗（如JAK抑制剂）的重要疗效评估标志物。

实验操作关键要点

WB优化（磷酸化与总蛋白同步检测）：

样本处理：细胞需用含磷酸酶抑制剂（PhosSTOP+Na₃VO₄） 的RIPA裂解液冰上裂解30分钟，14000g离心20分钟取上清；若检测激活状态，需同步设置IL-3刺激组（10ng/mL，30分钟） 作为阳性对照（STAT5磷酸化水平升高）；

电泳条件：8% SDS-PAGE凝胶（因分子量较大），恒压80V浓缩胶、120V分离胶，确保92/94kDa条带分离；

抗体孵育：一抗推荐1:2000稀释（5% BSA+TBST），4℃摇床孵育过夜，HRP标记二抗（山羊抗兔）1:5000室温孵育1小时，曝光时间控制在30-60秒避免信号饱和。

IHC-P注意事项（白血病骨髓样本）：

抗原修复：采用柠檬酸钠缓冲液（pH6.0） 微波修复20分钟（95℃），增强核内信号；

阳性对照：推荐使用慢性粒细胞白血病（CML）骨髓活检组织（白血病细胞胞核呈棕黄色阳性，胞质弱阳性）；

封闭处理：5%驴血清+0.3% Triton X-100室温封闭45分钟，配合3% H₂O₂阻断内源性过氧化物酶（15分钟）。

IP实验要点（蛋白相互作用研究）：

样本预处理：1mg细胞裂解液中加入3μg抗体，4℃旋转孵育4小时，Protein A/G磁珠（50μL）捕获免疫复合物；

洗脱条件：0.1M甘氨酸-HCl（pH2.8）室温洗脱10分钟，立即用1M Tris-HCl（pH8.0）中和，用于WB检测JAK2-STAT5复合物。
