产品介绍The protein encoded by this gene is a member of the STAT family of transcription factors. In response to cytokines and growth factors, STAT family members are phosphorylated by the receptor associated kinases, and then form homo- or heterodimers that translocate to the cell nucleus where they act as transcription activators. This protein is activated by, and mediates the responses of many cell ligands, such as IL2, IL3, IL7 GM-CSF, erythropoietin, thrombopoietin, and different growth hormones. Activation of this protein in myeloma and lymphoma associated with a TEL/JAK2 gene fusion is independent of cell stimulus and has been shown to be essential for the tumorigenesis. The mouse counterpart of this gene is found to induce the expression of BCL2L1/BCL-X(L), which suggests the antiapoptotic function of this gene in cells. [provided by RefSeq, Jul 2008]

白血病相关蛋白18/癌蛋白18（Stathmin/STMN1）是一种微管解聚蛋白，通过抑制微管聚合和解聚已形成的微管，参与细胞有丝分裂、迁移和增殖过程。其在急性白血病、乳腺癌、肺癌等多种肿瘤中高表达，通过促进肿瘤细胞快速分裂和侵袭转移，成为肿瘤预后不良的标志物，也是潜在的抗癌药物靶点。

实验操作关键要点

WB优化（白血病细胞样本）：

样本处理：白血病细胞（如K562）需用含磷酸酶抑制剂（如Na₃VO₄） 的RIPA裂解液冰上裂解30分钟，12000g离心15分钟取上清；

电泳条件：15% SDS-PAGE凝胶（因分子量小），恒压80V浓缩胶、120V分离胶，确保17kDa主条带清晰；

抗体孵育：一抗推荐1:1500稀释（5% BSA+TBST），4℃摇床孵育过夜，HRP标记二抗（山羊抗兔）1:5000室温孵育1小时。

IHC-P注意事项（肿瘤组织切片）：

抗原修复：采用柠檬酸钠缓冲液（pH6.0） 微波修复15分钟（95℃），增强胞质信号；

阳性对照：推荐使用急性髓系白血病骨髓活检组织（白血病细胞胞质呈棕黄色阳性）；

封闭处理：5%驴血清室温封闭30分钟，配合3% H₂O₂阻断内源性过氧化物酶（10分钟）。
