It encodes a ubiquitous cytosolic phosphoprotein proposed to function as an intracellular relay integrating regulatory signals of the cellular environment. The encoded protein is involved in the regulation of the microtubule filament system by destabilizing microtubules. It prevents assembly and promotes disassembly of microtubules. 靶点与功能

STMN1（Stathmin 1，又称LAP18/癌蛋白18）是一种微管解聚蛋白，通过结合微管蛋白二聚体抑制微管组装，参与调控细胞有丝分裂、迁移和增殖。其过度表达会导致细胞周期紊乱，在白血病、肺癌、乳腺癌等多种肿瘤中高表达，与肿瘤侵袭转移、化疗耐药密切相关，是肿瘤预后评估和靶向治疗的重要标志物。

实验操作关键要点

WB优化（白血病细胞系样本）：

样本处理：使用含磷酸酶抑制剂（如Na₃VO₄） 的RIPA裂解液冰上裂解30分钟，12000g离心15分钟取上清，避免STMN1磷酸化修饰丢失；

电泳条件：15% SDS-PAGE凝胶（因分子量小），恒压80V浓缩胶、120V分离胶，确保17kDa主条带清晰；

抗体孵育：一抗推荐1:1000稀释（5% BSA+TBST），4℃摇床孵育过夜，HRP标记二抗（山羊抗兔）1:5000室温孵育1小时。

IHC-P注意事项（白血病骨髓活检样本）：

抗原修复：采用柠檬酸钠缓冲液（pH6.0） 微波修复15分钟（95℃），增强胞质信号；

阳性对照：推荐使用急性髓系白血病（AML）骨髓切片（白血病细胞胞质呈棕黄色阳性）；

封闭处理：5%驴血清室温封闭30分钟，配合3% H₂O₂阻断内源性过氧化物酶（10分钟）。

IF双标实验（微管共定位研究）：

推荐与α-微管蛋白抗体共染，使用Alexa Fluor 488（绿色，标记STMN1）和594（红色，标记微管），激光共聚焦观察STMN1对微管网络的解聚效应。
