产品介绍This gene belongs to the stathmin family of genes. It encodes a ubiquitous cytosolic phosphoprotein proposed to function as an intracellular relay integrating regulatory signals of the cellular environment. The encoded protein is involved in the regulation of the microtubule filament system by destabilizing microtubules. It prevents assembly and promotes disassembly of microtubules. 靶点与功能

神经突触素1（Synapsin I）是一种突触前膜磷酸蛋白，通过结合突触小泡膜和细胞骨架（如微管、肌动蛋白）调控突触小泡的锚定、动员及神经递质释放，是突触形成、突触可塑性及神经信号传递的关键分子。其表达异常与阿尔茨海默病、癫痫、精神分裂症等神经疾病密切相关，也是神经内分泌肿瘤（如嗜铬细胞瘤）的重要诊断标志物。

实验操作关键要点

WB优化（脑组织/神经元样本）：

样本处理：使用含磷酸酶抑制剂（如PhosSTOP） 的RIPA裂解液冰上裂解30分钟，12000g离心15分钟取上清，避免磷酸化位点去修饰；

电泳条件：10% SDS-PAGE凝胶，恒压80V浓缩胶、120V分离胶，确保78kDa主条带与非磷酸化条带分离；

抗体孵育：一抗推荐1:2000稀释（5% BSA+TBST），4℃摇床孵育过夜，HRP标记二抗（山羊抗小鼠）1:5000室温孵育1小时。

IHC-P注意事项（突触定位研究）：

抗原修复：采用柠檬酸钠缓冲液（pH6.0） 微波修复15分钟（95℃），促进突触前膜抗原暴露；

阳性对照：推荐使用大鼠海马组织切片（锥体细胞层突触前膜呈棕黄色颗粒状阳性）；

封闭处理：5%羊血清+0.3% Triton X-100室温封闭45分钟，降低非特异性背景。

IF双标实验（突触结构观察）：

推荐与突触小泡蛋白（Synaptophysin） 共染，使用Alexa Fluor 488（绿色，标记Synapsin I）和594（红色，标记Synaptophysin），激光共聚焦观察突触前膜共定位。
